A common organization of the T-DNA genes expressed in plant hairy roots induced by different plasmids of Agrobacterium rhizogenes

Abstract: The recently described pathogenic plasmid from Agrobacterium rhizogenes 2659 induces "hairy root" proliferation in infected plants which synthesize a new opine type, provisionally called cucumopine. The T-DNA restriction site map of 2659 differs from other pRi T-DNAs, i.e., mannopine and agropine. However all these three different T-DNAs have a similar organization as detected through hybridization. This, previously shown by our cross-hybridization studies between mannopine and agropine pRi T-DNAs, is here ext… Show more

“…1) -were individually cloned in Bin 19 [22] and recombinant plasmids were transferred via triparental mating to the host strain LBA4404 [24]. Data from the only available transcript map [25] and, subsequently, from partial DNA sequencing (J. Brevet, personal communication) allowed us to assign the relative transcript(s) to the cloned T-DNA segments (Fig. 1).…”

“…Agropine and mannopine are also produced by some strains of A. tumefadens (Lahners et al 1984). Formtilae and/or names of a range of opines are given by Combard and Baucher (1988), Petit et al (19S3), Ryder et al (1984) and Szegedi et al (1988).…”

“…The TR-DNA of the agropine-type Ri plasmid contains genes that are homologous to the iaaM and iaaH genes of the Ti T-DNA (Huffman et al 1984). TL-DNA is considered to be essential for the induction of hairy roots, since cucumopine-and mannopine-type Ri plasmids have a single T-DNA that is homologous to the TL-DNA of the agropine-type Ri plasmid (Combard and Baucher 1988). Eighteen open reading frames (ORF) have been identified on the TL-DNA (Slightom et al 1986).…”
