A Combined Patch-Clamp and Electrorotation Study of the Voltage- and Frequency-Dependent Membrane Capacitance Caused by Structurally Dissimilar Lipophilic Anions

Abstract: Interactions of structurally dissimilar anionic compounds with the plasma membrane of HEK293 cells were analyzed by patch clamp and electrorotation. The combined approach provides complementary information on the lipophilicity, preferential affinity of the anions to the inner/outer membrane leaflet, adsorption depth and transmembrane mobility. The anionic species studied here included the well-known lipophilic anions dipicryl- no mobile charges could be detected by electrorotation, despite their strong membran… Show more

“…The modeling of cell and particle trapping was performed using COMSOL Multiphysics 5.3 and the add‐on AC/DC and Particle Tracing modules. Simulated particles were configured as single‐shell particles (composed of a cytoplasm surrounded by a thin membrane with human embryonic kidney (HEK) cell properties from the literature (rext=7.5μm,σcp=0.533S/m,εcp=71,Gcm=0S/m2,andCcm=8.5mF/m2) . The equation used to implement the single‐shell model with G cm and C cm in COMSOL Multiphysics are presented in the Supporting Information.…”

“…The hybrid probe hVOS employs FRET between membrane-anchored GFP and a dye, DPA, that partitions into the plasma membrane in a membrane voltage-dependent manner. While this hybrid probe produces the largest reported Δ F/F values among all genetically encoded voltage-sensitive probes, the high concentration of the lipophilic anion DPA has a significant effect on the activity of excitable membranes (Fernández et al, 1983; Chanda et al, 2005; Zimmermann et al, 2008). Thus reduced DPA concentrations were required to obtain action potential responses (Chanda et al, 2005) resulting in much smaller fractional changes (Δ F/F ).…”
