A Collagenous Cementum-Derived Attachment Protein Is a Marker for Progenitors of the Mineralized Tissue-Forming Cell Lineage of the Periodontal Ligament

Abstract: The periodontal ligament (PDL) is a fibrous and cellular connective tissue that mediates tooth attachment to bone, and it comprises fibroblastic and mineralized tissue-forming (MTF) progenitors. The MTF progenitors are believed to give rise to the cementoblastic and osteoblastic lineages. Cementum attachment protein (CAP) is a collagenous cementum-derived protein which binds strongly to osteoblasts, moderately to PDL cells, and weakly to gingival fibroblasts. The aim of the present study was to determine the r… Show more

“…Osteogenic, chondrogenic, and adipocytic lineages represented the mesodermal germ layer. Nodular mineralized deposits characteristic to human osteoblastic cultures derived from bone marrow stromal progenitor cells [30,41] were identified with Alizarin Red S staining in 5 weeks old cultures as described elsewhere [41] (Fig. 3A).…”

“…hOMSC were generated by explantation as described by us elsewhere [30] and expanded in low-glucose Dulbecco's modified Eagle's medium (LG-DMEM) and 10% fetal calf serum (FCS); (expansion medium). Human embryonic stem cells (hESCs) (HES-1 cell line [31]) with a stable normal karyotype (46XX) were maintained as described in [32] and served as positive controls in assays described below.…”

“…Approximately 42 % of cultured adult human periodontal ligament cells are clonogenic which could be expanded by over 222 doublings [33]; therefore, these clones could represent cultured stem cell population of the periodontal ligament. About 20 % of these putative stem cells do not differentiate into mineralized tissue in the presence of dexamethasone unless they were first induced with BMP 2 [34].…”
