A Coding Variant of ANO10, Affecting Volume Regulation of Macrophages, Is Associated with Borrelia Seropositivity

Abstract: In a first genome-wide association study (GWAS) approach to anti-Borrelia seropositivity, we identified two significant single nucleotide polymorphisms (SNPs) (rs17850869, P = 4.17E-09; rs41289586, P = 7.18E-08). Both markers, located on chromosomes 16 and 3, respectively, are within or close to genes previously connected to spinocerebellar ataxia. The risk SNP rs41289586 represents a missense variant (R263H) of anoctamin 10 (ANO10), a member of a protein family encoding Cl -channels and phospholipid scram-

“…Notably, we also found higher caspase 3/7 activity in cells expressing ANO10, and even more remarkably, coexpression of both LRRC8A and ANO10 potentiated caspase activity. These results confirm the roles of LRRC8A and ANO10 in apoptotic cell death and support the notion of a functional interaction of LRRC8A and ANO10 (Hammer et al 2015). We found earlier that ANO10 induces VRAC when expressed in oocytes from X. laevis, while the variant R263H fails to induce VRAC (Hammer et al 2015).…”

We found earlier that ANO10 induces VRAC when expressed in oocytes from X. laevis, while the variant R263H fails to induce VRAC (Hammer et al 2015). Notably, expression of LRRC8A also induced VRAC in Xenopus oocytes, which is very different from mammalian cells, where overexpression of LRRC8A did not generate additional VRAC (Qiu et al 2014;Voss et al 2014).…”
