A class of sulfonamides as carbonic anhydrase I and II inhibitors

Abstract: Four groups of novel sulfonamide derivatives: (i) acetoxybenzamide, (ii) triacetoxybenzamide, (iii) hydroxybenzamide and (iv) trihydroxybenzamide, all having thiazole, pyrimidine, pyridine, isoxazole and thiadiazole moieties were prepared and their inhibitory effects were studied on two metalloenzymes, i.e. carbonic anhydrase isozymes (hCA I and II), purified from human erythrocyte cells by Sepharose-4B-l-tyrosine-sulfanilamide affinity chromatography. These enzymes are present in almost all living organisms t… Show more

“…CAs have a variety of functions, including carbon dioxide (CO 2 ) transport, pH adjustment, ion transport, gastric acid formation, bone resorption, calcification and genesis, development, and tumor invasion (27,28). As one isoenzyme of CA, CA2 is widely distributed in vivo and has the highest activity.…”

“…They interfere with PABA (p‐aminobenzoic acid) in the biosynthesis of tetrahydrofolic acid, which is a basic growth factor essential for the metabolic process of bacteria. Therefore, sulfonamides are an important class of medicinally active compounds, and they are used as preventive and chemotherapeutic agents against various diseases . Sulfonamides possess many types of biological activities such as antibacterial, anti‐carbonic anhydrase, hypoglycemic, diuretic, antithyroid, anti‐hypertensive, antiviral, antiprotozoal, antifungal, anti‐inflammatory, and antitumor activity .…”

“…Carbonic anhydrase functions in many physiological and pathological processes, such as transport, bone resorption, calcification, and tumorigenesis are demonstrated. Thus, 16 different isozymes that differ from each other with tissue distribution, cell localization, catalytic activity, and resistance to inhibitors have been described . CAI and CAII antibodies have been identified in various autoimmune and idiopathic diseases; Grave's disease, metabolic syndrome, polycystic ovary syndrome, acute anterior uveitis, and Behcet's disease .…”
