A CAPS marker to assist selection of tomato spotted wilt virus (TSWV) resistance in pepper

Abstract: The hypersensitive resistance to tomato spotted wilt virus (TSWV) in pepper is determined by a single dominant gene (resistant allele: Tsw) in several Capsicum chinense genotypes. In order to facilitate the selection for this resistance, four RAPD (among 250 10-mer primers tested) were found linked to the Tsw locus using the bulked segregant analysis and 153 F2 individuals. A close RAPD marker was converted into a codominant cleaved amplified polymorphic sequence (CAPS) using specific PCR primers and restricti… Show more

“…The Tsw gene has been mapped in the distal portion of chromosome 10 [136]. A CAPS marker (SCAC 568) tightly linked 0.9 cM away to the Tsw locus has been identified in a segregant F 2 population C. chinense (PI152225) × C. frutescens (PI195301) [137]. The resistance conferred by the Tsw gene is overcome by high temperatures (28-33 • C) and early plant virus inoculations (two-to four-true-leaf stages) [138].…”

“…In the Capsicum species, several linkage maps have been reported (Prince et al 1993, Lefebvre et al 1995, Livingstone et al 1999, Kang et al 2001. Thus, DNA markers linked to various agronomic characters were developed (Caranta et al 1999, Moury et al 2000, Blum et al 2002, Matsunaga et al 2003, Sugita et al 2004. These DNA markers are now available for MAS for some traits.…”

“…Furthermore, DNA markers linked to various useful characteristics have been identified. These markers include disease resistance to pepper mild mottle virus (PMMoV; L 4 ) (Matsunaga et al 2003), cucumber mosaic virus (Ben et al 2001a), tomato spotted wilt virus (Moury et al 2000) and potato virus Y (Caranta et al 1999), fruit shape (Ben et al 2001b), pungency (Blum et al 2002) and mature fruit color (Popovsky andParan 2000, Huh et al 2001). Moreover, we have developed SCAR markers linked to PMMoV resistance (L 3 ) (Sugita et al 2004).…”
