A Brief Review on the Regulatory Roles of MicroRNAs in Cystic Diseases and Their Use as Potential Biomarkers

Abstract: miRNAs are small endogenous conserved non-coding RNA molecules that regulate post-transcriptional gene expression through mRNA degradation or translational inhibition, modulating nearly 60% of human genes. Cystic diseases are characterized by the presence of abnormal fluid-filled sacs in the body, and though most cysts are benign, they can grow inside tumors and turn malignant. Recent evidence has revealed that the aberrant expression of a number of miRNAs present in extracellular fluids, including plasma or s… Show more

“…Dysfunctional granulosa cells (GCs), characterized by a poor apoptosis and increased proliferation, have proved to be typical of PCOS and are usually associated with impaired ovulation and folliculogenesis [ 11 ]. Elevated levels of circulating insulin could be responsible for GC abnormalities: specifically, it is able to favor the overexpression of mir-93 and the downregulation of mir-145, which in turn enhance cell growth and inhibit apoptosis [ 12 , 13 ].…”

“…The pharmacological modulation of miR-145 can thus represent a therapeutic strategy to resolve GCs dysfunction in PCOS [ 12 ]. Furthermore, human GCs have been found to present significant lower levels of miR-126-5p and miR-29a-5p, along with the overexpression of its target, the klotho protein, which has been suggested to regulate GC survival/apoptosis by modulating the insulin/IGF-1, Wnt1 and Akt signaling pathways [ 11 , 89 ].…”

“…It has also been stated that a single transcript might be targeted by multiple miRNAs, while a sole miRNA can regulate up to 200 different mRNAs ( Ruiz-Manriquez et al, 2022c ; Iacomino, 2023 ). The investigation of how miRNA-mediated regulation is influenced by factors such as miRNA abundance/localization, cell type, and cell state is currently being extensively studied ( Vishnoi and Rani, 2023 ); however, it has been hypothesized that an aberrant expression of these molecules influences critical biological processes leading to various pathological outcomes associated with diseases ( Diener et al, 2022 ).…”
