A brain-derived neurotrophic factor (BDNF) haplotype is associated with antidepressant treatment outcome in mood disorders

Abstract: Brain-derived neurotrophic factor (BDNF) has been studied extensively in relation to the susceptibility to mood disorders (MD), although it remains to be clarified whether BDNF is a susceptibility locus for MD phenotypes, including therapeutic response to antidepressants. We have performed a single-marker and haplotype association study of eight TagSNPs polymorphisms in the genomic region containing the BDNF gene in 342 control subjects and 374 patients with MD, and have tested the association with antidepress… Show more

“…A similar heterosis effect was also reported for another SNP within BDNF (rs90887) [182]. Preliminary findings were provided for other BDNF SNPs (rs90887, rs61888800, rs7124442 and rs11030104) [179,182,183]. A strong association was established between rs962369 and an increase in suicidal ideation during antidepressant treatment [117].…”

“…A functional variant at codon 66, resulting in a valanine to methionine change (Val66Met), is reported to correspond with drug response. Carriers of the Met allele were reported to have better treatment outcomes (Gratacos et al, 2008;Kato & Serretti, 2010), however, others did not find any correlation between the Val66Met variant and treatment response (Kato & Serretti, 2010;Tsai et al, 2003;Wilkie et al, 2007). Furthermore, genetic susceptibility to depression was not associated with the BDNF Val166Met variant (Gratacos et al, 2007;Lopez-Leon et al, 2008).…”
