A bioengineered peripheral nerve construct using aligned peptide amphiphile nanofibers

Abstract: Peripheral nerve injuries can result in lifelong disability. Primary coaptation is the treatment of choice when the gap between transected nerve ends is short. Long nerve gaps seen in more complex injuries often require autologous nerve grafts or nerve conduits implemented into the repair. Nerve grafts, however, cause morbidity and functional loss at donor sites, which are limited in number. Nerve conduits, in turn, lack an internal scaffold to support and guide axonal regeneration, resulting in decreased effi… Show more

“…Lastly, bioactive peptides for epitope-specific biological interactions can also be incorporated; for example, cell adhesion molecules are often incorporated to mimic native extracellular matrices (Figure 8). In recent years, the majority of in vivo applications using cylindrical PA-based scaffolds include regeneration for, bone [198], nerves [199202], and reproductive organs [203]. …”

“…The use of IKVAV-PA to examine the ability of Schwann cells to attach and proliferate within the material was reported. The results show that Schwann cells adhere and proliferate better on IKVAV-PA as compared to non-modified PA. 61 Stupp et al continue their work on the development of stimuli-responsive dynamic materials. The ability of materials to respond to changes in physical structure and chemical composition would greatly improve the development of synthetic materials that ultimately mimic the dynamic features of the ECM.…”
