A bifunctional nanomodulator for boosting CpG-mediated cancer immunotherapy

Abstract: Unmethylated cytosine-phosphate-guanine (CpG) oligonucleotides (ODNs) possess high immunostimulatory activity and represent attractive tools for cancer treatment. However, their success in eliminating large solid tumors was hampered by the immunosuppressive tumor microenvironment. Herein, we report that the design of a novel MnO-CpG-silver nanoclusters (AgNCs)-doxorubicin (DOX) conjugate for enhanced cancer immunotherapy, in which MnO nanosheets function as unique supports to integrate the chemotherapy drug DO… Show more

“…For example, MnO 2 NSs were designed as a unique support to integrate DOX and cytosine‐phosphate‐guanine (CpG)‐Ag NPs together for TME‐mediated DOX and CpG deliveries . Another case is that core–shell Au@MnO 2 (AM) NPs were produced for FL/PA/MR‐imaging guided O 2 ‐boosted PDT and immunotherapy by Cai group ( Figure a,b) .…”

“…In this treatment, CuS nanoparticles transform the laser energy to the thermal energy to induce the cancer cell apoptosis, while CpG/LPS act as the bioadjuvant to stimulate the immunity against the tumors. Similarly, doxorubicin was encapsulated in MnO 2 ‐CpG‐silver nanoclusters via π–π stacking interactions, which significantly inhibited the growth of large solid tumors …”

“…To avoid the undesired in vivo distribution of activated immune cells, nanomaterials, including liposomal, polymeric, or inorganic nanocarriers, have been developed to concentrate immunostimulatory drugs within tumors to boost the regional immunity . Several pioneering works have demonstrated the potential to stimulate the local T‐cell immune response by delivering immunomodulators with various types of carriers to achieve effective tumor elimination . Although great efforts have been devoted to enhancing the stability and the targeting efficiency of the delivered molecules, little has been investigated on the precise control of the interactions between immune cells and the cancer cells, which ultimately determines the therapeutic efficacy .…”
