A 42-Kilodalton Annexin-Like Protein Is Associated with Plant Vacuoles

Abstract: A 42-kD, calcium-dependent, membrane-binding protein (VCaB42) was associated with partially purified vacuole membranes. Membrane-dissociation assays indicated that VCaB42 binding to vacuole membranes was selective for calcium over other cations and that 50% of VCaB42 remained membrane bound at 61 2 11 nM free calcium. A 13-amino acid sequence obtained from VCaB42 showed 85% similarity with the endonexin fold, a sequence found in the annexin family of proteins that is thought to be essential for calcium and lip… Show more

“…We have also compared calcium-binding activity (on ligand blots) with a vacuolar annexin protein, VCaB42 Randall, 1994, 1997). Although the vacuolar annexin binds calcium in the native state, with an apparent dissociation constant (K d ) of 60 nm (Seals and Randall, 1994), it does not bind calcium by the calcium ligand-blot assay (Fig. 8).…”

“…VCaB45 binds calcium in the ligand blot but the vacuole annexin, VCaB42, does not. VCaB42 was obtained as previously described (Seals and Randall, 1994) and loaded in the first lane of each gel. VCaB45 was eluted from a nitrocellulose spot cut from a two-dimensional gel.…”

“…On the other hand, there are annexins (like Drosophila annexin IX and X, Hydra annexin XII) that appear to be specific for the individual species (Johnston et al, 1990;Schlaepfer et al, 1992). There are also indications for the existence of unique plant-specific annexins (Seals et al, 1994;Smallwood et al, 1990). However, annexins do not seem entirely ubiquitous as annexinlike coding sequences have not been identified in the Saccharomyces cerevisiae genome, and they have not been found in protozoa and prokaryotes.…”

“…However, at the present stage any conclusion about the subcellular localization of CR proteins in spinach leaves would be premature. Although in plant cells the vacuole, together with the cell wall (Gilroy et al, 1993;Seals et al, 1994), is generally regarded to be the main site of accumulation of Ca 2+ , there is equal evidence that the ER may play an important role in Ca 2+ homeostasis (Bush et al, 1989. ER-enriched microsomal fractions from celery were shown to contain otherwise uncharacterized Ca 2+ -binding proteins with apparent molecular masses of 55 and 58 kD (Randall, 1992).…”
