Goniothalamus marcanii Craib (synonym of Goniothalamus tamirensis Pierre ex Finet & Gagnep), is locally known as Khao Lam in Thai. The previous study showed that the isolated compounds from stem barks exhibited high cytotoxicity to human tumor cell lines. The objective of the research was to investigate compounds which showed cytotoxic effect. Column chromatography was used for isolation. The structures of the compounds were investigated by spectroscopic methods. The bioactivity of the compounds was determined by SRB assay. The result showed that four compounds, a styryl-lactone: (+)-goniodiol (1); and three alkaloids: ouregidione (2); noraristolodione (3); velutinam (4) were isolated from G. marcanii stem barks. Compound 3 possessed the strongest activity (EC 50 35.56 and 50.75 μM), for A549 (lung) cells and HeLa (cervix) cells, respectively. Compound 1-3 displayed cytotoxic activity but less than that of doxorubicin, a conventional chemotherapeutic drug, the EC 50 levels of which were 0.13 and 0.12 μM for A549 and HeLa cells, respectively. On the other hand, compound 2 and 4 which could be evaluated from selective index values (SI 1.25 and 1.70, respectively) showed lower toxicity towards normal cells (MRC5) than doxorubicin (SI = 1.07). From this study, G. marcanii stem barks found to be a new source of compounds 1-4. In addition, the investigation of compound 3 in cytotoxic activity and the evaluation of compounds 1 and 2 for cytotoxic activity to HeLa cell line have been reported for the first time.

Release of a payload in a spatiotemporal fashion has a substantial impact on increasing therapeutic efficacy. In this work, a novel monolayer of gold nanoparticles (AuNPs) featuring light-responsive ligands was investigated as a potential drug carrier whose drug release can be triggered by UV light. Hydrophobic molecules were noncovalently entrapped in the compartments of its monolayers. Once irradiated with UV light, the dinitrobenzyl linker was cleaved, leading to release of the entrapped agent. AuNPs were characterized using UV spectrophotometry, TEM, and a zetasizer. A naturally occurring compound extracted from Goniothalamus elegans Ast was chosen as a hydrophobic model drug. Entrapment and release of dye were monitored using fluorimetry. The percent encapsulation of dye was of 13.53%. Entrapped dye can be released upon UV irradiation and can be regulated by changing irradiation time. Up to 83.95 ± 2.2% entrapped dye can be released after irradiation for 20 minutes. In the absence of UV light, dye release was only 19.75%. For comparison purposes, AuNPs having no dinitrobenzyl groups showed a minimal release of 12.23% and 11.69% with and without UV light, respectively. This demonstrated an alternative strategy to encapsulate drugs using a noncovalent approach followed by their controlled release upon UV irradiation.

The genus Goniothalamus belongs to the Annonaceae family. The pure compounds from Goniothalamus rongklanus and Goniothalamus latestigma were isolated by repeated column chromatography. The structures were recognized by NMR spectral methods. The chemical compounds of ethyl acetate extract (EtOAc) from the stems of G. rongklanus were identified as goniotriol (1) and stigmasterol glucoside (2). The EtOAc leaves extract of G. latestigma yielded a compound, which was identified as pinocembrin (3). The ethyl acetate (EtOAc) and methanol (MeOH) extract were tested for anti-HIV-1 RT and cytotoxic activities against P-388, KB, HT 29, MCF-7, A 549, ASK and CL cell lines. The EtOAc and MeOH extract of G. rongklanus showed evidence of anti-HIV-1 RT inhibition at 76.44 and 88.48 %, respectively. The EtOAc extract of G. rongklanus and G. latestigma showed cytotoxic activities on KB and HT 29 with an ED 50 at <4 µg/mL. In addition, antibacterial study on extracts and isolated compounds was also performed. Antibacterial study was evaluated using nine strains (Staphylococcus aureus, Enterobacter aerogenes, Escherichia coli 0157: H7, Escherichia coli (ETEC), Escherichia coli (EPEC), Proteus mirabilis, Salmonella typhimuriam, Shigella flexneri and Vibrio cholera) by Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC) method. Goniotriol was the most effective antibacterial with a MIC in the range <0.16 -0.6 mg/mL and MBC in the range 0.3 ->5 mg/mL. The antibacterial activity of goniotriol has been reported for the first time.
