Untitled

Abstract: Background: Several cell lines and primary cultures benefit from the use of positively charged extracellular matrix proteins or polymers that enhance their ability to attach to culture plates. Polyethyleneimine is a positively charged polymer that has gained recent attention as a transfection reagent. A less known use of this cationic polymer as an attachment factor was explored with several cell lines.

“…3b, no hemolysis of red blood cells was caused by the PDs, even at a concentration of 4000 μg/mL, indicating favorable blood compatibility. Thus, the cellular experiments and hemolysis assay confirmed the biocompatibilities of the PDs, especially in comparison with PEI alone, the cytotoxicity of which has been cited as a significant concern in the literature26.…”

“…We tested the cytocompatibility of PELD using an established method for polycations with the fibroblast cell line, NIH 3T3 [24,25]. We compared PELD to PEI, a common polycation used in cell culture and as a polymer transfectant for non-viral gene delivery [26,27]. We also used PEAD as a positive control, as we have previously shown it to be highly biocompatible both in vitro and in vivo [17].…”

“…Cells were transiently transfected with polyethyleneamine as described48. N2a cells with PGRN deletion were generated by infecting N2a cells with lentivirus expressing Cas9 only or Cas9 and guide RNA targeted to mouse Grn (oligos with 5′-CACCGCGGACCCCGACGCAGGTAGG-3′ and 5′-AAACCCTACCTGCGTCGGGGTCCGC-3′ were ligated to pLenti-CRISPR (Addgene)).…”
