Background: Severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) pandemic is a serious health problem all over the world including Egypt, thus realizing the predictive factors and disease's characteristics is an essential issue. Objectives: To evaluate the characteristics of laboratory-confirmed cases of SARS-CoV-2 infection in Fayoum governorate, Egypt, and to determine the predictive factors of disease severity. Methods: One hundred-fourty patients confirmed with SARS-CoV-2 from the Fayoum governorate, Egypt, were collected in this descriptive multicenter study. The subtype classification of SARS-CoV-2 was according to the World Health Organization (WHO) guideline SARS-CoV-2 disease severity classification. Patients were divided into a asymptomatic/non-severe cases group and a severe/critical case group. Each patient was subjected to chest computed tomography (CT), clinical, and laboratory assessment in form of complete blood count, neutrophil-to-lymphocyte ratio (NLR), lymphocyte-tomonocyte ratio (LMR), platelet-to-lymphocyte ratio (PLR), liver function tests, urea, creatinine, C-reactive protein (CRP), serum ferritin, and D-dimer. Results: Severe/critical patients were older (52.0±12.6) with a statistical significantly higher rate of diabetes mellitus, hypertension, and tuberculosis (TB) (p<0.001) than non-severe cases. Dyspnea was the most prevalent significant symptom among severe /critical group (87.5%, p<0.001). A negative correlation between radiographic score and oxygenation index (r = -0.302, p=0.007). Using the ROC analysis, the area under curve (AUC) was highest for a radiographic score, D-dimer, CRP, ferritin, and NLR, with p<0.05 in severe/critical cases. Conclusions: Elevated CRP, D-dimer, serum ferritin, radiograph score, and NLR may contribute to the judgment of SARS-CoV-2 severity, and help clinicians to evaluate the patient's condition. Co-infection of SARS-CoV-2 and TB can occur, and may progress towards severe SARS-CoV-2.

Identification of a stripe rust resistance gene YrSF in Chinese wheat (Triticum aestivum) landrace Sifangmai

Wheat stripe rust is an important disease and can seriously threaten wheat production. The most economical, safe and effective approach to manage this disease is to breed and deploy resistant varieties. Sifangmai is a Chinese wheat landrace and maintained adequate resistance to the Chinese stripe rust races. In order to decipher genetic bases of resistance in Sifangmai, the F1, F2 and F2:3 of the cross Sifangmai/Taichung 29 were tested at adult stage with race CYR32. Genetic analysis revealed that resistance in Sifangmai was controlled by a single dominant gene named as YrSF. Six SSR on chromosome 5BS showed polymorphism among ‘Taichung 29’, Sifangmai, and resistant and susceptible bulks. Chinese Spring nulli‐tetrasomic and monosomic lines were confirmed YrSF on chromosome 5B. A linkage map of YrSF was constructed and the closely linked marker to YrSF was Xgpw8015 with 1.8 cM. Based on the comparisons of resistance infection type, pedigree analysis, gene origin and chromosomal position of previously reported Yr genes on chromosome 5B, YrSF is different from these known genes and is likely a novel gene.

The low-molecular-weight glutenin subunits (LMW-GS) with extra cysteine numbers have attracted great research interest for their potential quality value. In this study, 14 LMW-i type genes (YD1-YD14) were isolated from three types of Chinese wheat landraces; and nine of 14 genes (YD1-YD9) had nine cysteines, and the other five genes contained eight cysteines. Phylogenic analysis suggested that all 14 LMW-i genes were related to Glu-A3-1 variants Glu-A3-17/FJ 549934 and Glu-A3-15/FJ 549932. Six randomly selected genes, five genes including YD1 with nine cysteines and the remaining one with eight cysteines, were successfully expressed in bacteria as mature proteins with a molecular mass of ~ 46 kDa. These proteins were traced to corresponding seed storage proteins for having similar elution times in reverse phase high-performance liquid chromatography (RP-HPLC) profiles. Mass spectrometry verified that bacterial expressed protein pET-30a-YD1 was LMW-i. Dough mixing experiments for incorporation of 50 mg pET-30a-YD1 proteins into the base flour of weak gluten wheat cv. "Chuannong 16" indicated that the dough strength of mixing flours was noticeably weaker than that of the control, which was reflected by mixing parameters in 8-min curve width, peak width, peak height, mixing time, and right of peak slope. The results suggested that the LMW-i genes with nine cysteine residues in the present study contributed to inferior quality properties for wheat flour.
