Somatic Embryogenesis: Fundamental Aspects and Applications

Somatic embryogenesis is a very useful micropropagation technique, due to its high multiplicative capacity that offers the potential for large-scale propagation using temporary immersion bioreactors. The temporary immersion system is mainly based on the contact of plant tissue with culture medium by certain cycles of immersion, avoiding the problems of hyperhydricity, malformed embryos, and low conversion rates, which occur in continuous immersion systems. The automation of some or all of the phases of the process of somatic embryogenesis in a bioreactor could reduce labor and gellant costs and increase micropropagation efficiency, allowing high-quality plantlets to be obtained through more efficient and controlled protocols. This chapter describes the different types of temporary immersion bioreactors that have been used to increase or scale somatic embryogenesis in different plant species.

The present study describes a protocol for plant regeneration via somatic embryogenesis in temporary immersion system (TIS) for Camptotheca acuminata. Somatic embryos were induced by culturing hypocotyl segments from 14-day-old in vitro grown C. acuminata seedlings in TIS. Hypocotyl segments were placed in culture vessels modified with a mechanical device to support the fixation of explants. Cultures were maintained under a 16 h photoperiod with a light intensity of 60 lmol m -2 s -1 PPF at 25 ± 1°C. After 16 weeks of incubation embryogenic calli were formed above the edge of the mechanical device in the basal Murashige and Skoog (MS) medium containing 35 g l -1 sucrose and without hormonal supplementation. For plantlet regeneration, somatic embryos at cotyledonary stage were cultured in three different concentrations of 6-benzylamino-purine (0.5, 1.0 and 1.5 mg l -1 BAP) and in plant growth regulator (PGR) free medium. In general, 0.5 mg l -1 BAP was found to be the most effective concentration for growth and development of Camptotheca embryos in TIS. Conversion of somatic embryos into plantlets was also successfully achieved on sterile substrates moistened with 0.5 mg l -1 BAP. Plantlets derived from cotyledonary embryos were rooted in vitro with 0.5 mg l -1 indole-3-butyric acid (IBA) before transfer to ex vitro conditions.

Temporary immersion bioreactors (TIBs) are being used to propagate superior plant species on a commercial scale. We demonstrate a new TIB design, a Hydrostatic-driven TIB (Hy-TIB), where periodic raising and lowering the media reservoir maintains the advantages of temporary immersion of plant tissues without requiring large amounts of gas to move the media that is a characteristic of other TIB designs. The advantage of utilizing low volumes of gas mixtures (that are more expensive than air) is shown by a doubling of the growth rate of plant root cultures under elevated (40%) oxygen in air, and with CO2 supplementation showing improved phototrophic and photomixotrophic growth of seedless watermelon meristem cultures. The development of this bioreactor system involved overcoming contamination issues associated with utilizing very low gas flow rates and included utilizing microchip pressure sensors to diagnose unexpected changes in internal bioreactor pressure (± 20 Pa ∼0.0002 atm) caused by flexing of non-rigid plastic bag vessels. The overall design seeks to achieve versatility, scalability and minimum cost such that bioreactor technology can play an increasing role in the critical need to improve plant productivity in the face of increasing demand for food, reduced resources, and environmental degradation. © 2016 American Institute of Chemical Engineers Biotechnol.
