IntroductionDendrobium species have been widely used for many health disorders since ancient times. However, due to unrelenting collection to meet the increasing demand for their use in medication and other health products, the natural habitats of medicinal Dendrobium transparens have been devastated and are on the verge of extinction.MethodsAn efficient in-vitro propagation protocol for Dendrobium transparens using seed derived protocorms was established and genetic homogeneity of the in-vitro regenerants and the wild plant was studied. ResultsThe maximum seed germination was observed in Full strength Murashige and Skoog medium (FMS). Induction of protocorms were achieved on basal as well as half-strength MS medium. The highest number of shoot (11.9 shoots/explant) was achieved in half MS medium fortified with 100 mL/L coconut water in addition with Benzyl amino purine (BAP) 1 mg/L and Kinetin 2 mg/L. Further, elongated shoots were transferred to full and half strength MS root initiating medium supplemented with different concentration of auxins. However, a maximum of (8.3 ± 0.6, 4.9 ± 0.1 cm) roots were achieved in full MS medium fortified with 100 mL/L coconut water and Napthalene acetic acid (NAA) 1.5 mg/L. Ten rapid Random Amplified Polymorphic DNA (RAPD) and Inter Simple Sequence Repeats (ISSR) primers were used to analyze genetic stability among in-vitro and mother plant. RAPD primers produced a total of 23 fragments while ISSR primers produced a total of 16 fragments. ConclusionThe amplified bands of all the samples of in-vitro plants were similar to bands of mother plant. The present research reported here is indicating the applicability of tissue culture for true-to-type plant production and conservation of D. transperens.

Dendrobium chryseum is an endangered epiphytic and lithophytic orchid species having medicinal and ornamental value. The objective of the present study is to develop micropropagation protocol for D. chryseum. The immature seeds of D. chryseum were used as an explant for the micropropagation. Protocorms were developed within 90 days in Murashige and Skoog (MS) medium without plant growth regulators. Protocorms were differentiated into micro shoots after 30 days of transfer to MS medium supplemented with 2 mg/L Benzyl amino purine (BAP), 1 mg/L kinetin and 10% coconut water. Among eleven different combinations of plant growth regulators (BAP, á-Naphthalene acetic acid (NAA), Adenine sulphate, Kinetin and coconut water) treated for shoot multiplication, maximum number of shoots were obtained in 0.5 mg/L BAP + 0.5 mg/L NAA (5.8 ± 0.53 SE shoots per explant). Longest shoot length was recorded in MS medium with Adenine sulphate (1 mg/L) (2.54 ± 0.03 cm SE). Root induction was carried out by using MS medium with different concentrations (0.5, 1, 1.5, 2.0 mg/L) of Indole butyric acid (IBA) and NAA. The highest numbers of roots and longest root length, both were observed at 2 mg/L IBA (4.63 ± 0.56 SE and 2.09 ± 0.25 cm SE respectively). 2 mg/L NAA showed poor response for root number (0.94 ± 0.21 SE) and root length (0.43 ± 0.07 cm SE). Successful acclimatization of in-vitro grown plantlets was done by wrapping the plantlet with moss kept on fine pine bark and the survival rate of plantlet was about 80% after 30 days. This protocol could be helpful for the effective mass propagation and ex situ conservation of D. chryseum.

Bulbophyllum auricomum Lindl. is a rare orchid and has flowers with an attractive fragrance. The present study investigated the tissue culture method for micropropagation. Capsules derived from artificial self-pollination were obtained for the best seed germination in MS basal medium. Plant growth regulators (1.0 mg•L -1 of BAP and 2.0 mg•L -1 of NAA) were affected by callus induction from subcultured pseudobulb explants. For the callus subculture, different natural plant extracts were tested in 11 treatment media. Among them, MS medium with 150 mL•L -1 of coconut water was generally effective in fresh weight (1.75 ± 0.08) and (3.01 ± 0.20) of callus proliferation and PLBs induction at 1 and 2 months, respectively, followed by an MS combination of 30 g•L -1 of banana and 20 g•L -1 of potato extract. The results of a comparative study of different MS mediums containing plant growth regulators with a natural extract combination and MS medium supplemented with natural extract only showed that MS medium supplemented with a combination of natural extracts (150 mL•L -1 of coconut water) and plant growth regulators (2.0 mg•L -1 of BAP and 1.0 mg•L -1 of NAA) obtained the highest shoot regeneration (3.37 ± 0.17) and (6.41 ± 0.68) after 1 month and 2 months of culturing, respectively.
