Actinidia, the kiwifruit is approximately 20 to 26 million years old. It is known widely for its dioecious nature; hermaphroditism has also been found to exist in its male inconstant type. Floral development in this plant species progresses through seven unique stages, following the conventional (A) BCE floral model. Male parts of the flower develop early during the third stage, whereas, the degeneration of these parts (pollens) in female plants occurs during later stages owing to programmed cell death. After exploring the sex-linked segment of the kiwifruit genome, the researchers have determined that the Y-encoded sex-determinant genes Shy Girl (SyGl) and Friendly Boy (FrBy) act independently as the suppressors of feminization and promoters of male factors, respectively. Non-expression of SyGl in males results in the formation of hermaphrodites; yet, masculine behaviour has been reported to develop in this fruit crop with plunking of FrBy gene into the female plants. These findings have created new avenues for generating horticulturally important self-pollinating hermaphrodite vines, eliminating the need to waste orchard space on non-fruiting male varieties.

Kiwifruit belong to the genus Actinidia with 54 species apparently all functionally dioecious. The sex-determinants of the type XX/XY, with male heterogametic, operate independently of the ploidy level. Recently, the SyGI protein has been described as the suppressor of female development. In the present study, we exploited the CRISPR/Cas9 technology by targeting two different sites in the SyGI gene in order to induce a stable gene knock-out in two tetraploid male accessions of Actinidia chinensis var. chinensis. The two genotypes showed a regenerative efficiency of 58% and 73%, respectively. Despite not yet being able to verify the phenotypic effects on the flower structure, due to the long time required by tissue-cultured kiwifruit plants to flower, we obtained two regenerated lines showing near fixation of a unique modification in their genome, resulting in both cases in the onset of a premature stop codon, which induces the putative gene knock-out. Evaluation of gRNA1 locus for both regenerated plantlets resulted in co-amplification of a minor variant differing from the target region for a single nucleotide. A genomic duplication of the region in proximity of the Y genomic region could be postulated.

RESUMO Este trabalho faz uma revisão dos estágios de aquisição da escrita à luz da Linguística Aplicada, e fornece uma alternativa diversa dos conhecidos estágios ferreirianos, muito utilizados pelo professor alfabetizador. Para tal, lanço mão de achados e conclusões fornecidos pelos estudos de aquisição da escrita, de orientação fonológica (ABAURRE, 1998, 1999; CAGLIARI, 1992a, 1992b, 1998), bem como de estudos de aquisição da linguagem que evidenciam o papel do molde silábico no processo aquisicional (McNEILAGE; DAVIS, 1990). São utilizados, como corpora, os registros da pesquisa de campo e experimental realizada ao longo do ano letivo de 2006, com 20 crianças de escolas públicas da cidade de Salvador, em cinco encontros periódicos individuais de produção escrita. Tais produções me permitem concluir que o percurso da aquisição da escrita inicia-se com a ausência de relação grafo-fônica, passando por uma série de etapas cíclicas de relações som/letra, até atingirem as relações ortográficas, cuja dependência som/grafia é superada.
