Abstract: Significant bioavailabilities of CGP 64128A were achieved following subcutaneous, intra-peritoneal and intra-tracheal administration. Pulmonary delivery represents a promising mode of non-parenteral dosing for antisense oligonucleotides. The dose-dependent increase in pulmonary bioavailability suggests that low doses may be retained in the lungs for local effects whereas higher doses may be suitable for the treatment of a broader spectrum of systemic diseases.

“…Similar results were obtained when 30 μ g or 10 μ g of RNA were fed to mice (Table S2). The small percentage of dietary RNA detected indicates a low level of uptake, and is consistent with other recent works (Nicklin et al 1998; Snow et al 2013). …”

“…Thus, a subtype-specific inhibitor would be required for the functional analysis of oatps. It is known that AONs are distributed mainly to the liver and kidney, less so to skeletal muscles, and not at all to the brain ( , ). For the analysis of oatps in the liver and kidney, subtype-specific oatp ENA AONs, such as OA1-9, OA2-13c, and OA3-2c, which were designed and constructed in this study, might be useful in vivo .…”

“…Thus, the systemic absorption of orally ingested RNA is negligible. For a 20mer DNA oligonucleotide, constructed with stabilizing phosphorothionate linkages to prevent degradation in the gastrointestinal tract, oral bioavailability in rats was at best 0.3% after gavage administration (Nicklin et al, 1998). Kendal D. Hirschi and colleagues reported on an unusual sRNA from herbs, flowers and vegetables with a comparably high stability in gastrointestinal fluids (Yang et al, 2016(Yang et al, , 2018.…”
