Abstract: Using our gp120/41-expressing, NK cell activity-resistant CEM.NKR cell clones as targets in HIV-1-specific antibody-dependent cellular cytotoxicity (ADCC) assays, we demonstrate here that the serum titers of anti-HIV-1 ADCC antibodies bear a significant (P < 0.05) positive correlation with the peripheral blood CD4+ T cell counts and a negative one with the number of copies of HIV-1 RNA in the plasma of HIV-infected individuals. These findings underscore the importance of these antibodies as a protective immune… Show more

“…Second, it is unknown if these HIV-specific IgG1 antibodies are involved in control of viral replication. While effector function has correlated with nonprogression in numerous studies and in both humans and macaques (12,13,20), whether this association is causative remains to be determined.…”

“…The sum of these and other functions can be measured more generally as antibody-dependent cellular viral inhibition (ADCVI), and a number of studies have suggested that these extraneutralizing effector functions of antibodies play a role in an effective immune response to HIV (summarized in reference 18). In both humans and macaques, ADCC activity has correlated with delayed progression to AIDS (12,13,19,20), and ADCVI has been reported in early stages of simian immunodeficiency virus (SIV) infection, coincident with the appearance of anti-gp140 antibodies. Viruses that had mutated to escape neutralization remained susceptible to ADCVI during later infection, and ADCVI was correlated with control of viral replication, showing that at least in this model, ADCVI played a substantial role in a protective immune response (21).…”

“…ADCC responses are detectable in plasma shortly after the resolution of acute viremia and correlate inversely with disease progression (14)(15)(16)(17)(18)(19)(20). Greater ADCC responses have also been observed in individuals who exhibit elite control of HIV-1 in the absence of antiretroviral therapy (21,22).…”

“…For instance, viral escape has been reported for ADCCtargeted epitopes, suggesting the existence of ADCC-related immune pressure on the virus (40). Some but not all studies have correlated ADCC responses with lower viremia (41)(42)(43), and HIV controllers display elevated ADCC activity (44,45). Regarding the virus itself, the accessory proteins Vpu and Nef decrease ADCC mediated by some monoclonal or polyclonal antibodies, likely by limiting the amount of Env at the surface of infected cells (24,(46)(47)(48)(49)(50)(51).…”
