Abstract: Our findings suggest that perfluorohexyloctane is tolerated in the rabbit eye for 9 weeks. Since avascular rabbit retina is more susceptible to mechanical and toxic damage than vascularized primate retina, we suggest proceeding from here, and investigating tolerance and quality of tamponade of perfluorohexyl octane in man for up to 9 weeks.

“…Zeana et al [5] investigated the intra-ocular tolerance of F 6 H 8 in vitrectomized eyes of pigmented rabbits on slit lamp biomicroscopy, fluorescein angiography and electroretinography. After enucleation, light and electron microscopies were performed.…”

“…Perfluorohexyloctane (F 6 H 8 ) has a density of 1.35 g/cm 3 . Its biocompatibility has been demonstrated in the experimental animal [5] and in human eyes [6, 7]. It was tolerated without any obvious signs of damage to the retina or to the optic disc with an average duration of 76 days [7].…”

“…They were the first intraocular tamponades used beyond surgical time [41]. In addition, it has been demonstrated that SFAs can be successfully used also as intraoperative tools to unfold and lay down the retina.…”

“…F6H8 was well tolerated for three months in rabbit eyes [11], and it was introduced initially as a solvent for silicone oil to remove silicone oil remnants from intraocular surfaces [64, 65]. Further, F6H8 was investigated as an intraoperative tool and as a long-term tamponade in several small case series [12, 66].…”

“…A third group of substances, liquid semifluorinated alkanes (SFAs), appeared to have the potential to act as heavy internal tamponade agents [10]; in particular, perfluorohexyloctane (F6H8) seemed to be well tolerated in long-term animal studies [11]. In clinical practice, the use of F6H8 provided adequate reattachment rates and few signs of retinal damage; however, it was associated with a high rate of postsurgical inflammation and an early rate of emulsification of droplets into the entire eye [12].…”
