9G DNAChip: microarray based on the multiple interactions of 9 consecutive guanines

Abstract: We introduce the phenomenon of molecular recognition to immobilize oligonucleotides on AMCA slides for the production of 9G DNAChips. Facile and efficient method for the immobilization of the oligonucleotides appended with consecutive nine guanine bases is described. The 9G DNAChips shows more than 90% hybridization efficiency at 25 °C in 30 min.

“…Before hybridization, the 9G HPV DNA chips were covered with Secure-Seal hybridization chambers. An immobilization density of 6.2 pmol/cm 2 was calculated for the probes immobilized on the HPV 9G DNA chip according to the method in our previous report (17).…”

“…Recently reported 9G (GGGGGGGGG) DNA chips based on 9G DNA chip technology show high SBR (PMT gain ϭ 48%) and 100% target-specific hybridization, with more than 90% hybridization efficiency at 25°C in 30 min (17,18 …”

“…Among them, 9G DNA chip technology is based on the phenomenon of molecular recognition to immobilize oligonucleotides for the production of DNA chips. The 9G probes are immobilized by multiple interactions of the nine consecutive guanines on the aminomethylcoumarin acetate (AMCA) monolayer [14]. The HPV 9G DNA chip test, which has been approved by the Korea Food and Drug Administration (KFDA), displays a promising diagnostic characteristic of 100% clinical sensitivity and specificity, making it a promising diagnostic tool for HPV genotyping [1518].…”

“…HPV 9G DNA genotyping was performed according to the manufacturer's protocol [1415] under the supervision of Dr. A. Lee (a pathologist with a specialty in gynecopathology and molecular pathology). Briefly, all specimens of swabs were placed in a SurePath preservative vial (BD Diagnostics-Tripath BD Biosciences, Oxford, UK) and transferred to the pathology laboratory.…”

“…The MTB-DR-RIF 9G test is a new diagnostic tool based on the recently reported 9G technology [20], and the controller DNA technology (CDT) [21]. The MTB-DR-RIF 9G test can discriminate multiple SNPs in the five codons (c511, c516, c522, c526, and c531) of MTB strains at 25 C, in less than 30 min after PCR.…”
