8-Bromo-3′5′ -Adenosine Monophosphate Stimulates the Endocrine Activity of Human Cytotrophoblasts in Culture*

Abstract: Cytotrophoblasts, purified from human term placentae, were cultured in the absence or presence of 8-bromo-cAMP or 8-bromo-cGMP. 8-Bromo-cAMP provoked a dose-dependent increase in the secretion of hCG and progesterone within 24 h. After 48 h, hCG secretion increased by more than 200-fold, and progesterone secretion increased nearly 5-fold. 8-Bromo-cGMP had no effect on hCG secretion. In culture in serum-supplemented medium, the mononuclear cytotrophoblasts aggregated and fused to form syncytia. This morphologic… Show more

“…This enhanced the expression of genes representative of syncytiotrophoblast endocrine activity (Kao et al 1992). It is also reported that treatment with 8-bromo-cAMP causes cytotrophoblasts isolated from term placenta to aggregate within hours and an increase in hCG and progestin secretion in serum-supplemented medium (Feinman et al 1986). Moreover, Wice et al (1990) showed that BeWo cells underwent fusion and morphological differentiation in standard medium after treatment with effectors of cAMP metabolism.…”

“…aggregate to form syncytia, in spite of a significant increase in hCG secretion induced by 8-bromo-cAMP in a serumfree medium (Feinman et al 1986). These results suggest that the functional differentiation of trophoblast induced by 8-bromo-cAMP is not accompanied by morphological differentiation under serum-free conditions.…”

“…However, except cAMP/protein kinase A (PKA) signaling pathway, its regulatory mechanism has not been definitively characterized. Activation of the cAMP signaling increases ERVWE1 gene expression (Frendo et al 2003, Chen et al 2008 and can induce syncytia formation in primary cytotrophoblasts and human choriocarcinoma BeWo cells (Feinman et al 1986, Mi et al 2000. Activation of the cAMP/PKA signaling pathway in turn increases the expression of a transcription factor, glial cell missing factor homolog 1 (GCM1, previously called hGCMa), which binds to the upstream region of the ERVWE1 gene and then stimulates ERVWE1 gene transcription in a cellspecific manner (Yu et al 2002, Chang et al 2005.…”

“…Progesterone can be synthesized from cholesterol by the syncytiotrophoblast (Tuckey 2005). A variety of endogenous factors such as estrogen, HCG, and cyclic AMP are known to regulate placental progesterone production (Feinman et al 1986, Rodway et al 1988, Chaudhary et al 1992. Thus, further study of the mechanism by which the hormone network controls progesterone production in placenta is a prerequisite to gain insight into the mechanism of pregnancy and parturition.…”
