730 Histotripsy focused ultrasound ablation induces immunological cell death in treated and distant untreated tumors

Abstract: BackgroundHistotripsy, a novel image-guided, robotically assisted sonic therapy platform, is a non-invasive and non-thermal tumor ablation modality. We have previously shown that histotripsy potentiates profound innate and adaptive anti-tumor responses in addition to direct tumor destruction.1 In this study, we sought to characterize the biomarkers of tumor cell death pathways immediately after histotripsy and after the induction of adaptive anti-tumor immune responses in preclinical settings.MethodsImmunocomp… Show more

“…Sonodynamic therapy has been widely applied as an effective and noninvasive cancer therapeutics. , Ultrasound can destroy tumor and generate tumor fragments, which can be used as antigens to form in situ vaccines and induce the systemic immune responses . Sonodynamic therapy can induce ICD to elicit effective immune responses and achieve better therapeutic results in combination with immunotherapy . Besides, ultrasound can promote endosomal escape of mRNA to facilitate efficient translation and improve therapeutic efficacy .…”

“…Tumor ablation is the destruction of a tumor through temporary extreme conditions (i.e., heat, cold, chemicals) and is an attractive low-cost alternative for primary tumor control when surgery is impractical. Additionally, several types of ablations have been shown to elicit an anti-tumor immune response through the release of tumor-associated antigens (TAAs) following tumor necrosis [ 13 , 14 ]; thus, systemic control could be achieved by a local ablation with a robust induction of the immune response. Tumor ablation paired with checkpoint inhibitors is currently being studied in many active clinical trials with varied success; however, checkpoint inhibitors remain cost prohibitive.…”
