7‐Dehydrocholesterol Encapsulated Polymeric Nanoparticles As a Radiation‐Responsive Sensitizer for Enhancing Radiation Therapy

Abstract: Therapeutics that can be activated by radiation in situ to enhance the efficacy of radiotherapy are highly desirable. Herein, 7‐Dehydrocholesterol (7‐DHC), a biosynthetic precursor of cholesterol, as a radiosensitizer, exploiting its ability to propagate the free radical chain reaction is explored. The studies show that 7‐DHC can react with radiation‐induced reactive oxygen species and in turn promote lipid peroxidation, double‐strand breaks, and mitochondrial damage in cancer cells. For efficient delivery, 7‐… Show more

“…While the incubation time and NP concentration do impact biocompatibility, cell viability did not decrease below 60% even at the highest concentration of 100 μg/mL. The improved biocompatibility upon 7-DH loading could be attributed to the drug's antioxidant activity on cells 63 due to its nonspecific release, as shown in Figure 5.…”

“…Compared to cells treated with ionizing radiation (IR) alone, 7-DHC@PLGA NPs associated with IR-treated cells demonstrated an increase of caspase-3 activity of 24.3% and a decreased count of all colonies formed under test radiation doses. In addition, the cell viability percentage at the dosage of two was 0.209 for 7-DHC@PLGA NPs combined with IR, whereas it was 0.530 for a single IR representing the dose-modifying factor of 2.536 ( Delahunty et al, 2022 ). “…They are converted in the liver and kidney into the active form: 1, 25-dihydroxy vitamin D (calcitriol) [5,6]. Additionally, vitamin D metabolism occurs in other tissues, and the active form serves an autocrine/paracrine function [7,8]. Serum 1, 25-dihydroxy vitamin D should be higher than 30 ng/mL [9] to maintain a healthy and normal vitamin D level.…”
