6S RNA modulates growth and antibiotic production in Streptomyces coelicolor

Abstract: The aim of this study was to contribute to clarifying the role of 6S RNA in the development and control of antibiotic biosynthesis in Streptomyces coelicolor. Due to the low energetic cost of gene silencing via 6S RNA, it is an easy and rapid means of down-regulating the expression of specific genes in response to signals from changes in the environment. The expression of 6S RNA in S. coelicolor is not constitutive, and its accumulation is adapted to changes in nutritional conditions. The 6S RNA of S. coelicol… Show more

“…In many organisms, 6S RNA-RNA polymerase complexes have been detected by co-immunoprecipitation or in vitro analysis (e.g. B. subtilis (14, 35, 70), L. pneumophila (29), C. burnetii (53), Aquifex aeolicus (71), and Streptomyces coelicolor (41)) supporting a general similarity to Ec 6S RNA. However, detailed understanding of interactions in other organisms is likely to require biochemical or structural analysis to uncover any potential differences from Ec 6S RNA.…”

“…altered timing of sporulation and cell density changes in stationary phase) are associated specifically with the loss of Bs 6S-1 RNA (34, 36). Streptomyces coelicolor 6S RNA also accumulates with a profile similar to Ec 6S RNA and influences growth rate (41). …”

“…The 6S sRNA of S. coelicolor is capable of interacting with RNA polymerase and is a template for the transcription of short oligonucleotides. Knowledge about expression and function of 6S sRNA from antibiotic-producing Streptomyces has been limited until recently, but the deletion and complementation of the ssrS demonstrated its important role in the regulation of growth and expression of key genes for the biosynthesis of actinorhodin (Mikulik et al, 2014). These studies demonstrate the possibility of metabolic engineering of Streptomyces using ncRNAs.…”

“…Confirmation of the expected employment of small RNAs in the regulation of cell processes, including primary metabolism, developmental transitions, antibiotic production, and various stress responses is being increasingly reported (Palecková et al, 2007 ; Pánek et al, 2008 ; Swiercz et al, 2008 ; D'Alia et al, 2010 ; Mikulík et al, 2014 ). Hundreds of cis- acting asRNAs were identified using RNA sequencing in two recent studies (Vockenhuber et al, 2011 ; Moody et al, 2013 ).…”
