6-Gingerol-rich fraction prevents disruption of histomorphometry and marker enzymes of testicular function in carbendazim-treated rats

Abstract: Previous investigations demonstrated that 6-gingerol-rich fraction (6-GRF) prevented testicular toxicity via inhibition of oxidative stress and endocrine disruption in CBZ-treated rats. The influence of 6-GRF on alterations in histomorphometry and marker enzymes of testicular function in CBZ-treated rats which hitherto has not been reported was investigated in this study. The animals were orally administered either CBZ (50 mg/kg) alone or in combination with 6-GRF (50, 100 and 200 mg/kg) for 14 consecutive day… Show more

“…63 Activities of free lysosomal enzymes have been shown to increase when testicular steroidogenesis is elevated. 64 However, a reduction in the ACP activity, as observed in DOX-treated rats, indicates decreased testicular steroidogenesis, and this may be correlated with reduced secretion of gonadotrophins. ALP is involved in the biosynthesis of nucleic acids, nuclear proteins, phospholipids, cleavage of phosphate esters, mobilizing carbohydrates and lipid metabolites for spermatozoa utilization.…”

“…These enzymes are acknowledged indices of testicular function commonly assessed in reproductive toxicology. [ 40 ] Testicular ALP hydrolyses 6‐phosphoglucose, an intermediate in the glycolytic pathway, to free glucose which is essential for the proliferation and differentiation of spermatogenic cells. Testicular ACP activity is largely linked to functionality and survival of spermatogonial stem cell.…”

“…The activity of glucose‐6‐phosphate dehydrogenase (G6PD) was analyzed as previously described. [ 40 ] Lactate dehydrogenase‐X (LDH‐X) was analyzed using an established procedure. [ 41 ] Acid phosphatase (ACP) activity was assayed in agreement with the established method [ 27 ] whereas alkaline phosphatase (ALP) activity was assayed as previously described.…”

“…Briefly, the testicular activity of glucose-6-phosphate dehydrogenase (G6PD) was assayed using nicotinamide adenine dinucleotide phosphate and glucose-6-phosphate as substrates as earlier reported. 41,42 Testicular acid phosphatase (ACP) and alkaline phosphatase (ALP) activities were assayed following established practice based on the degradation of p-nitrophenyl-phosphate in acid and alkaline milieu, respectively. 43,44 Also, testicular lactate dehydrogenase-X (LDH-X) activity was assessed by a standard technique that depended on the inter-change of lactate and pyruvate.…”
