53BP1 Enforces Distinct Pre- and Post-resection Blocks on Homologous Recombination

Abstract: Highlights d 53BP1 S25A point mutation rescues lethality of BRCA1 D11 mice without restoring HR d 53BP1 S25A mutation uncouples 53BP1 end-blocking activities pre-and post-resection d RIF1/shieldin blocks BRCA1-independent loading of RAD51 onto single-strand DNA d Shieldin and PTIP associate with 53BP1 to regulate distinct end-resection pathways

“…In striking contrast to 53BP1-deficient somatic cells in which DSB resection is consistently and acutely increased 18,30,33 , we found that resection lengths were comparable at all meiotic hotspots in 53bp1 -/and WT spermatocytes ( Fig. 4e).…”

“…We therefore performed END-seq on 53bp1 -/and BRCA1-deficient spermatocytes and measured overall resection lengths. In striking contrast to 53BP1-deficient somatic cells in which DSB resection is consistently and acutely increased 18,31,34 , we found that resection lengths were comparable at all meiotic hotspots in 53bp1 -/and WT spermatocytes ( Fig. 2d).…”

“…Rif1 interacts with 53BP1 and the Shieldin complex to process double strand DNA breaks, which affects the choice between homologous recombination and NHEJ. This choice plays a major role in immunoglobulin class switch recombination, and thus loss of Rif1 has dramatic consequences for immunoglobulin rearrangements (Silverman et al, 2004;Chapman et al, 2013;Callen et al, 2020) Notably, while many of these Rif1 functions in homologous recombination and NHEJ are conserved across phyla (Mattarocci et al, 2017;Fontana et al, 2018) and (reviewed in (Mattarocci et al, 2016)), the role of Rif1 in telomere length regulation is not conserved in mammals (Silverman et al, 2004;Xu and Blackburn, 2004;Buonomo et al, 2009). Rif1 regulates telomere length in a subset of species including S. pombe (Kanoh and Ishikawa, 2001) where it can interact with the telomere binding protein Taz1, and in C. glabrata (Castano et al, 2005) where it interacts with Rap1.…”
