5[prime] Proximal Regions of Arabidopsis Nitrate Reductase Genes Direct Nitrate-Induced Transcription in Transgenic Tobacco

Abstract: Nitrate redudase (NR) is the first enzyme in nitrate assimilation, a critica1 process for plant survival. l h e regulation of NR gene expression is complex, involving both interna1 and externa1 factors. O f these, nitrate indudion of NR gene expression has been studied most extensively and is well conserved among baderia, fungi, and higher plants. We are interested in understanding the mechanism of nitrate indudion of higher plant NR genes. Here we describe promoter analyses of the 5' flanking regions of the A… Show more

“…The NIT-2 motif has been widely found in genes for the enzymes involved in nitrogen metabolism (Marzluf, 1993). In higher plants these motifs have been found in the 5' flanking regions of the genes for NR (Lin et al, 1994) and NiR (Tanaka et al, 1994) in Arabidopsis thaliana, and for FNR in Oryza sativa (Aoki et al, 1995), as listed in Table I.…”

“…Deletion analysis of the 1.5-kb 5'-flanking regions of the Arabidopsis NIA1 and NIA2 genes showed that 238 and 188 bp, respectively, were important for nitrate induction of a reporter gene (CAT) in transgenic tobacco (Lin et al 1994). A 12 bp conserved sequence with a core consensus AG/CTCA, a "nitrate element", was necessary for nitrate dependent transcription (Hwang et al 1997).…”

“…Effects of NADK2 overexpression on GS/GOGAT pathway-related gene expression. A, Levels of GLN1;1, GLN1;3, GLN1;5, GS2, GLU1, GLT1, GDH1, and GDH2 transcripts in rosette leaves of 23- NIA and NIR expression (Pouteau et al, 1989;Lin et al, 1994;Scheible et al, 1997). In fact, NO 3 2 concentrations were approximately 10-fold higher in the nadk2 mutant (Fig.…”
