5-HT4 Receptors Constitutively Promote the Non-Amyloidogenic Pathway of APP Cleavage and Interact with ADAM10

Abstract: In addition to the amyloidogenic pathway, amyloid precursor protein (APP) can be cleaved by α-secretases, producing soluble and neuroprotective APP alpha (sAPPα) (nonamyloidogenic pathway) and thus preventing the generation of pathogenic amyloid-β. However, the mechanisms regulating APP cleavage by α-secretases remain poorly understood. Here, we showed that expression of serotonin type 4 receptors (5-HT 4 Rs) constitutively (without agonist stimulation) induced APP cleavage by the α-secretase ADAM10 and the re… Show more

“…Quantification of sAPP in culture supernatants by detection of alkaline phosphatase activity, as previously described (Cochet et al, 2013), showed that both treatments increased sAPP release and that 5-HT had the most prominent effect ( E max = 34 ± 5% of 5-HT maximal release for RS 67333; Figure 1A ). Then, the release of sAPPα was investigated in vivo in WT C57BL/6 mice after a single administration of RS 67333 (1 mg/kg, i.p.…”

“…The metalloproteinase region of the ADAMs family can degrade extracellular matrix and thus influence tumor progression [17, 18]. In this study, the effects of ADAM10-specific siRNAs on the migration capability and invasiveness of HepG2 cells are investigated.…”

“…When stimulated by 5-HT4 receptors agonists, however, sAPP α secretion is mediated through cAMP/Epac (exchange protein activated by cAMP) signalling. 57 A possible explanation for this study's findings is that α 2ARAs could directly activate α -secretase ADAM10 and sAPP α production through cAMP/Epac modulation.…”
