5-FU and ixabepilone modify the microRNA expression profiles in MDA-MB-453 triple-negative breast cancer cells

Abstract: This study aimed to discover new potential mechanisms of chemotherapy with drugs used in the treatment of luminal androgen receptor (LAR)-type triple-negative breast cancer (TNBC). We examined the microRNA (miRNA) expression profiles of LAR-type TNBC in vitro, and explored the variation in miRNA expression profiles in cells when treated with the chemotherapy drugs capecitabine and ixabepilone. The present study revealed that the expression levels of the three antitumor miRNAs, miR-122a, miR-145 and miR-205, we… Show more

“…From this perspective, is seems that miR-31-5p/3p could associated more probably with irinotecan then cetuximab response. Moreover, there is an increasing evidence describing link between chemotherapy and miRNAs regulatory network [ 10 , 11 ] indicating miRNAs involvement in chemosensitivity or chemoresistance [ 12 ]. On other hand, this evidence did not show any specific associations of miR-31-5p/3p and chemosensitivity, even in case of topoisomerase inhibitors like irinotecan (10–12).…”

“…As a commonly prescribed chemotherapy agents, its efficacy and safety have been proved in many clinical trials [24][25][26]. However, its inhibitory effect on bone tumor isn't often mentioned and the molecular mechanism behind this inhibition remains unexplored.…”

“…On the other side, in stromal cells, the expression of some cytokines is regulated by NF-κB, such as interleukin-1β (IL-1β), which can stimulate osteoclastogenesis and bone resorption in the presence of RANKL [7]; monocyte chemoattractant protein (MCP-1), which participates in unfavorable osteoclastogenesis and bone destruction [22]; and vascular endothelial cell growth factor (VEGF), which takes part in the process of angiogenesis [23]. , as a commonly used chemotherapy drug in clinical oncologic practice [24], is widely used for the treatment of various cancers, including gastrointestinal cancer, breast cancer, and neck cancer, and moreover, its efficacy and safety have been proved in many clinical trials [24][25][26]. It is well established that 5-FU targets a critical enzyme, thymidylate synthase (TS), which is the critical enzyme in DNA synthesis and repair [27].…”

“…vitro 5-fluorouracil declined levels of miRNA-221, miRNA-210, miRNA-21 and miRNA-10b in luminal androgen receptor-type triple-negative breast cancer MDA-MB-453 cell line [31]. Cytokines and growth factors mediate not only intercellular communication in the immune system, but also influenced on the other cell types [32][33].…”
