5-aminolevulinic acid-mediated photodynamic therapy can target aggressive adult T cell leukemia/lymphoma resistant to conventional chemotherapy

Abstract: Photodynamic therapy (PDT) is an emerging treatment for various solid cancers. We recently reported that tumor cell lines and patient specimens from adult T cell leukemia/lymphoma (ATL) are susceptible to specific cell death by visible light exposure after a short-term culture with 5-aminolevulinic acid, indicating that extracorporeal photopheresis could eradicate hematological tumor cells circulating in peripheral blood. As a bridge from basic research to clinical trial of PDT for hematological malignancies, … Show more

“…PDT using ALA has been reported to be effective against squamous carcinoma and basal cell carcinomas. Similarly, PDT was also found to be potent against cutaneous T-cell lymphoma (Sando et al, 2020). Photosensitizer such as chlorin 6 and photofrins showed anticancer activity against esophageal and lung carcinomas (Qiu, Kim, Penjweini, & Zhu, 2016;Srdanovi c et al, 2018).…”

“…7 More importantly, 1 O 2 photoactivation is only effective in cells with a high ALA intake. 8 However, it is essential to recognize that oxidative stress promotes a high production of reactive oxygen species (ROS), triggering various cell death pathways where the dominant mechanism depends on the ALA localization.…”

“…Such finding is due to the high density of ASGP receptors on the surface of liver cancer cells, which can bind BODIPY NPs containing galactose residues and be successfully internalized into Huh-7 cells via receptor recognition 48 , 49 , which is consistent with our previous report 24 . This result demonstrates that BODIPY PSs with versatile functional groups can have an improved targeting ability compared to imaging-guided PDT agents which cannot be chemically modified such as 5-aminolevulinic acid (ALA) 50 .
