4F‐MDMB‐BINACA: A New Synthetic Cannabinoid Widely Implicated in Forensic Casework

Abstract: This is the first report regarding the characterization of the new synthetic cannabinoid 4F‐MDMB‐BINACA. 4F‐MDMB‐BINACA was first analytically confirmed in seized drug material using gas chromatography–mass spectrometry (GC‐MS), liquid chromatography–quadrupole time‐of‐flight mass spectrometry (LC‐QTOF), and nuclear magnetic resonance (NMR) spectroscopy. Subsequent to this characterization, 4F‐MDMB‐BINACA was detected in biological specimens collected as part of forensically relevant casework, including medico… Show more

“…Similar trends have been reported in the United States demonstrating a globalized market in SCRA production and export. Krotulski et al described the first detection of 4F‐MDMB‐BINACA (7) in the United States in seized herbal material in December 2018 and noted the substance was first also detected in November 2018 in Singapore . Between November 2018 and March 2019, 4F‐MDMB‐BINACA (7) was detected in 29 toxicology cases.…”

“…The metabolism of APP‐BINACA has not been previously reported. Theoretical metabolites of APP‐BINACA were formulated based on knowledge of previously reported metabolic pathways for similar synthetic cannabinoids . The resulting theoretical metabolites were converted to formulae (and exact mass) and added to a targeted screening program.…”

“…LC‐QTOF‐MS data independent acquisition (DIA) allowed for detection of all theoretical masses and fragment ions during the run (within 100–550 Da). This afforded the opportunity to data mine for metabolites; a process that has been used successfully in the investigation of other members of this NPS class . The above formulae for APP‐BINACA metabolites were converted to exact mass and searched in the datafiles of APP‐BINACA positive extracts using PeakView® (SCIEX, Version 2.2) and MasterView™ (SCIEX, Version 1.1).…”
