Abstract: The results of this study indicate that the determining factor which alters the pharmacokinetic properties of these lipid carriers after intratumoral injection is not the rate of transfer from the interstitial space to the vascular side but the rate of intratumoral transfer from the injection site to the well-vascularized region.

“…To investigate the effect of size or charge on distribution, neutral liposomes (120-nm in diameter), and cationic liposomes (125-nm in diameter) were prepared. 24) Each formulation was labeled with [ 3 H]cholesteryl hexadecyl ether. The zeta potentials of neutral liposomes and cationic liposomes were Ϫ5.4 and 47.6 mV.…”

“…CPT was incorporated with a higher AE of 96.5%. According to the literature [30], the mean diameter of SLN seems to be suitable for reaching cancer cells, to be taken up and to release drug inside cells. In relation to the charge surface, unloaded and CPT-loaded SLN had slightly negative charge, and these values of ZP are good indications also for brain tumour targeting [30,31].…”

“…43) Furthermore, the complex formation of pDNA with cationic liposomes limits the diffusion within tissues due to the increased size and net charge. 33,44) Therefore, pDNA locally injected into tissues, such as muscle and skin, may only transfect cells around the injection site, which makes in vivo gene transfer tissue-specific.…”

“…When complexed with pDNA, cationic liposomes reduced the spread of pDNA in tumors following direct injection into the tissues. 44) Although the disposition of locally injected pDNA can be partially improved by using polyvinyl pyrrolidone, 119) hyaluronidase, 64,120) or electroporation, 90,121) these effects are limited to areas adjacent to the injection site.…”
