The biomolecular events resulting from the progression of hepatoblastoma remain to be elucidated. Fourier‐transform infrared (FTIR) and Raman spectroscopies are capable of noninvasively and accurately capturing the biochemical properties of biological tissue from its pathological status. Our aim was to probe critial biomolecular changes of liver accompanying the progression of pure foetal hepatoblastoma (PFH) by FTIR and Raman spectroscopies. Herein, biochemical alterations were both evident in the FTIR spectra (regions of 3100‐2800 cm−1 and 1800‐900 cm−1) and the Raman spectra (region of 1800‐400 cm−1) among normal, borderline and malignant liver tissues. Compared with normal tissues, the ratios of protein‐to‐lipid, α‐helix‐to‐β‐sheet, RNA‐to‐DNA, CH3 methyl‐to‐CH2 methylene, glucose‐to‐phospholipids, and unsaturated‐to‐saturated lipids intensities were significantly higher in malignant tissues, while the ratios of RNA‐to‐Amide II, DNA‐to‐Amide II, glycogen‐to‐cholesterol and Amide I‐to‐Amide II intensities were remarkably lower. These biochemical alterations in the transition from normal to malignant have profound implications not only for cyto‐pathological classification but also for molecular understanding of PFH progression. The successive changes of the spectral characteristics have been shown to be consistent with the development of PFH, indicating that FTIR and Raman spectroscopies are excellent tools to interrogate the biochemical features of different grades of PFH.

In this work, a novel biosensor was fabricated for detection of DNA damage induced by 4-nonylphenol (NP) and also determination of NP. To achieve this goal, a glassy carbon electrode (GCE) was modified with chitosan (Chit), gold nanoparticles (Au NPs) and DNA-multiwalled carbon nanotubes (DNA-MWCNTs). Then, the DNA-MWCNTs/ Au NPs/Chit/GCE was incubated with methylene blue (MB) to obtain MB-DNA-MWCNTs/Au NPs/Chit/GCE in which MB was used as the redox indicator. The modifications applied to the GCE were characterized by cyclic voltammetry (CV), electrochemical impedance spectroscopy (EIS), scanning electron microscopy (SEM), energy dispersive X-ray spectroscopic (EDS) and theoretical evidence. MB is a derivative of anthraquinone which can intercalate into double helix structure of DNA. By treating MB-DNA-MWCNTs/Au NPs/Chit/GCE with NP, a higher R ct was observed because the insertion of the NP may result in a more negative charge environment on the DNA surface which hinders accessibility of [Fe(CN) 6 ] 3-/4anion to the electrode surface. Change in the EIS response of the biosensor in the presence of NP was used to develop a novel system for monitoring the DNA damage induced by NP. The EIS technique was also used to develop a sensitive electroanalytical method for determination of NP.

Infrared (IR) spectroscopy is a powerful analytical technique that can be applied to investigate a wide range of biological materials (e.g., biofluids, cells, tissues), where a specific biochemical signature is obtained representing the 'fingerprint' signal of the sample being analysed. This chemical information can be used as an input data for classification models in order to distinguish or predict samples groups based on computational algorithms. One fundamental step towards building such computational models is sample selection, where a fraction of the samples measured during an experiment are used for building the classifier, whereas the remaining ones are used for evaluating the model classification performance. This protocol shows how sample selection can be performed in a computational environment (MATLAB) by using a combination of Euclidian-distance calculation and random selection, named Morais-Lima-Martin (MLM) algorithm, as a previous step before building classification models in biological-derived IR datasets.
