AbstractThe availability of appropriate propagation techniques is a major constraint to the domestication of the forest trees widely used by rural communities; such as Garcinia kola (Heckel). This study tested the ability of root and stem cuttings to regenerate vegetatively when treated with indole-3-butyric acid (IBA), and set in non-mist poly-propagator within a shaded nursery. It found that G. kola is amenable to propagation by softwood stem cuttings. Attention was given to the effect of cutting age (softwood, semi hardwood) and IBA application with regard to the sprouting and rooting efficiency. Bud and leaf emergence time were also investigated, as was the coppicing ability of the stump. Results revealed that roots and branches from mature forest trees did not sprout under any culture conditions. The highest percentages of rooting (70-85%) were obtained with softwood cuttings set in non-mist poly-propagator, regardless of hormonal treatment. The mean numbers of buds (2.9 ± 0.4) and roots (2.6 ± 0.1) produced by softwood cuttings was significantly greater than those obtained in the Control. The best average emergence time of buds (25.1 days ± 9.3) and leaves (36.8days ± 8.4) was obtained with cuttings treated with IBA and set in non-mist poly-propagator against, 54.4 ± 12.5 and 72.6 ± 3.4 days respectively for the Control. In general, non-mist poly-propagator gave the best propagation results. When coppiced, the shoots emerging from stumps with one node were the most vigorous.

Callusing and root induction in air layering was evaluated aiming at evolution of procedure for mass clonal propagation of mature ortets of five tropical broadleaf species differing in their potential for adventitious root formation in shoot cuttings as: Anogiessus latifolia < Boswellia serrata < Dalbergia latifolia < Gmelina arborea < Dalbergia sissoo. Two experiments were conducted in rainy season during consecutive years; without application of growth regulators in the first year and with growth regulators (T 1 -water, T 2 -100 ppm indole-3-acetic acid, T 3 -100 ppm thiamine-HCl and T4 -combination of T 2 + T 3 ) in the next year. Air layered branches were detached from the trees to record percentage of alive airlayers, callusing and rooting (%) as well as root number and root length. Response to air layering was found to be highly variable in five tree species but appeared to be feasible procedure for clonal propagation of mature ortets of B. serrata and D. sissoo with 100% (in auxin + thiamine treatment) and 83.3% (in auxin treatment) success, respectively. Maximum callusing (%) was found in D. latifolia while no callusing was observed in D. sissoo, which is most easy-to-root among all five species. Callus formation impedes adventitious rhizogenesis in air layers as significant negative correlation of callusing (%) and adventitious root formation was recorded in air layers of five tropical broadleaved tree species. Application of exogenous auxin alone or in combination with thiamine circumvents callusing to ensure direct development of roots for successful air layering.

The common method of propagation is through seeds but seed germination in Zanthoxylum armatum is very low due to the presence of hard seed coat, which might be a great hurdle for large scale production of plantlets. So an attempt was made in this study to see the effect of different growth hormones, their concentrations and different rooting media on the rooting and sprouting of Z. armatum. The stem cuttings of Z. armatum were treated with two types of auxins namely Indole-3-Butyric Acid (IBA) and Naphthalene Acetic Acid (NAA) at different concentrations (2000 ppm, 3000 ppm and 5000 ppm), while the untreated cuttings were used as control. The cuttings were planted in three different rooting media: sand, neopeat and mix (containing a mixture of sand, soil and vermin-compost). The completely randomized design was used for the experiment. The total number of stem cuttings of Z. armatum used in the experiment was 1080 for 18 treatments in three replicates (20 cuttings per treatment x 18 treatments x 3 replicates). The experiment was set up in controlled greenhouse conditions at Dabur Nepal Private Limited Nursery, Banepa, Kavre District. The parameters evaluated were root length, shoot length and number of roots per cutting. The collected data were analyzed statistically using R-program with Agricola. Least significant difference (LSD) and Duncan multiple Range Test (DMRT), as mean separation technique was applied to identify the most efficient treatment in the rooting and shooting behavior of Z. armatum (Gomez and Gomez, 1984). Hormone concentration and growth media significantly affected the rooting and shooting ability of Z. armatum stem cuttings. IBA was found to be more effective than NAA. Neopeat medium was better than sand and mix media. The highest number of roots (6.5) and root length (11.6 cm) were recorded under IBA 5000 ppm in neopeat medium.
