Transcriptome profiling analysis of the response to walnut polyphenol extract in <i>Helicobacter pylori</i>-infected cells

Abstract: Dietary intervention to prevent Helicobacter pylori (H. pylori) associated gastric diseases seems to be ideal with no risk of bacterial resistance, safe long term intervention, and correcting pathogenic mechanisms including rejuvenation of precancerous atrophic gastritis and anti mutagenesis. A transcriptome as set of all RNAs transcribed by certain tissues or cells demonstrates gene functions and reveals the molecular mechanism of specific biological processes against diseases. Here, we have performed RNAseq … Show more

“…All the assembled unigenes were searched against public protein databases, such as NCBI non-redundant protein sequences (NR) and Swiss-Prot for homology comparison. Only those matches with E-value below 10–5 were deemed significant ( Park et al, 2021 ; Jonathan et al, 2022 ). Furthermore, gene function prediction and classification were assigned based on the database of the Gene Ontology (GO) and Eukaryotic Orthologous Groups of proteins (KOG).…”

“…Radical scavenging activities were determined for these phenolic compounds. Additional transcriptomic profiling further ascertained how WPE can rescue gastric cells from H. pylori infection [78]. Multiple chemokinerelated pathways were altered by WPEs, including an array of anti-inflammatory and anti-oxidative genes, providing further evidence for the potential impact of walnut extracts on modulating key signaling pathways that may ultimately contribute to the carcinogenic process present in H. pylori-infected gastric cells [78].…”

Nunez-Sanchez et al [79] tested EA and its microbially-derived metabolites, the urolithins, at concentrations predicted in colon tissue following ingestion of EA-rich foods, based upon the established urolithin 'metabotypes' [52].…”

“…The inhibitory function of resveratrol and epicatechin on IL-8 is probably related to their inhibitory activity on the NF-κB pathway, which downregulates the expression level of IL-8. Research has found that Walnut polyphenol extracts prevent Hp -related tumor growth by inhibiting STAT3 phosphorylation and nuclear translocation in gastric mucosal cells [ 63 ], and reversing precancerous atrophic gastritis [ 64 ]. Nobiletin has been confirmed to inhibit Hp infection and prevent Hp -mediated GC [ 65 ].…”
