Molecular Characterization of All-Stage and Adult-Plant Resistance Loci Against Powdery Mildew in Winter Wheat Cultivar Liangxing 99 Using BSR-Seq Technology

Abstract: Winter wheat cultivar Liangxing 99, which carries gene Pm52, is resistant to powdery mildew at both seedling and adult plant stages. An F2:6 recombinant inbred line (RIL) population from cross Liangxing 99 × Zhongzuo 9504 was phenotyped with Blumeria graminis f. sp. tritici isolate Bgt27 at the adult plant stage in four field tests and the seedling stage in a greenhouse test. The analysis of bulk segregant RNA sequences identified an SNP-enriched locus, Qaprpm.caas.2B, on chromosome 2BL in the same genomic int… Show more

“…Our field resistance screening results showed that LX99 was highly resistant (immune) to S. mosellana, which was consistent with the findings in previous studies. LX99 has been demonstrated to be highly resistant to powdery mildew at both the seedling and adult plant stages. As LX99 is a high-quality wheat variety with disease and insect resistance, the understanding of its resistance mechanism to S. mosellana will contribute to the breeding of excellent wheat varieties.…”

“…Four QTL were responsible for the slow-mildewing resistance. Cultivars such as Liangxing 99, often have both ASR and APR (Zhao et al 2013;Shi et al 2021). The high-level resistance conferred by the ASR genes plays a signi cant extension role in the early stage of cultivar use.…”

“…BSA has been widely used in wheat genetic mapping, and it is one of the effective methods to rapidly obtain target genes [18,49,50]. For species with large and complex genomes like wheat, it is a huge and laborious project to complete genome sequencing.…”
