Sulfonamides complexed with metals as mycobacterial biofilms inhibitors

Abstract: Highlights Rapidly growing mycobacteria (RGM) causing infections by biofilm formation. Semi-quantitative method of biofilm formation was adapted for macrotechnics. Sulphonamides complexed with metals is a promising anti-adhesion agent. Sulphonamides complexed with metal is a possible inhibitor of signaling of biofilm formation.

“…Treatment of M. fortuitum is challenging because it has been reported to have high resistance to several drugs, including macrolides, aminoglycosides, and many antituberculosis drugs 18. However, M. fortuitum is almost always susceptible to sulfonamides, which in this case was successful in treating this patient’s infection 19…”

“…Coordination of the metal ion by the N atom from the heterocycle and the O atom of the -SO 2 NH-group, occurring in the case of Ru(III)-STZ and Ru(III)-SMZ, is one of the known modes of complexations in such systems [33]. Sulfonamides however can coordinate with metal ions in different ways [30,[33][34][35][36][37]. There could be found in the literature complexes with metal ions (e.g., Cu (II)) in which the coordination of the ion occurs only through the N atom of the thiadiazole ring [34], so the sulfonamide ligand, acts as a monodentate because neither the oxygen atom nor the nitrogen from the -SO 2 NHgroup are involved in coordination.…”

“…Interestingly, AB-2-29 is not the first example of a metal complex displaying antibiofilm activity against M. abscessus . Indeed, metal complexes made of gold, silver, copper or cadmium-containing sulfonamides have recently been shown to display similar properties against a variety of NTM [ 46 , 47 ]. Given the relatively high concentration of zinc, magnesium, calcium and iron found in the sputum of persons with CF and non-CF bronchiectasis [ 48 ], this observation provides a basis for the development of innovative therapeutics and adjunct therapeutics directed against difficult-to-cure NTM pulmonary infections.…”
