Apomorphine Targets the Pleiotropic Bacterial Regulator Hfq

Abstract: Hfq is a bacterial regulator with key roles in gene expression. The protein notably regulates translation efficiency and RNA decay in Gram-negative bacteria, thanks to its binding to small regulatory noncoding RNAs. This property is of primary importance for bacterial adaptation and survival in hosts. Small RNAs and Hfq are, for instance, involved in the response to antibiotics. Previous work has shown that the E. coli Hfq C-terminal region (Hfq-CTR) self-assembles into an amyloid structure. It was also demons… Show more

“…Note that the use of larger liposomes could result in light scattering and for this reason, we used liposomes of 100 nm. For polymerized Hfq-CTR, the amyloid signature is found at ~220 nm ( Figure 3 ), as expected [ 25 , 49 , 50 ]. Then we added liposomes (EPE) and observed that the amyloid signature disappeared.…”

“…These reasons may explain a possible low level of heliotrine in the whole extract, and thus, a decreased potency against Gram-negative bacteria in the present study. Apomorphine was found to be bactericidal against E. coli by targeting the pleiotropic bacterial regulator Hfq [ 21 ]. Pinoresinol showed good antibacterial activities against most of bacteria studied in the current work [ 22 ], while propiconazole and mebendazole demonstrated their potency against several fungi strains including Cryptococcus neoformans and Sclerotium rolfsii [ 23, 24 ].…”

“…Due to the diversity of their targets, sRNAs and Hfq are involved in numerous bacterial pathways, including virulence and pathogenicity [ 15 , 16 ]. For this reason, Hfq has attracted considerable attention [ 17 , 18 , 19 ].…”
