CircRNA-Mediated Regulation of Angiogenesis: A New Chapter in Cancer Biology

Abstract: Angiogenesis is necessary for carcinoma progression and is regulated by a variety of pro- and anti-angiogenesis factors. CircRNAs are RNA molecules that do not have a 5’-cap or a 3’-polyA tail and are involved in a variety of biological functions. While circRNA-mediated regulation of tumor angiogenesis has received much attention, the detailed biological regulatory mechanism remains unclear. In this review, we investigated circRNAs in tumor angiogenesis from multiple perspectives, including its upstream and do… Show more

“…2B ). As shown in previous studies, the lack of a 5′ cap and a 3′ polyadenylated tail provided circRNAs with more stability than linear mRNA [ 11 ]. Next, we treated PANC-1 and MIA-PaCa2 cells with RNase R, and evaluated degradation via RT-qPCR.…”

“…Circular RNAs (circRNAs), a class of novel noncoding RNAs, were discovered in 1976. CircRNAs derive from non-canonical splices of pre-mRNAs, and form a covalently closed loop structure lacking 5′ caps and 3′ tails [ 11 ]. CircRNAs present higher stability and resistance to exonuclease compared to the parental linear RNAs [ 12 ].…”

“…Angiogenesis: Angiogenesis-dependent tumor growth is an important characteristic of cancers[ 96 ]. Vascular endothelial GF (VEGF) and hypoxia-inducible factor-1α (HIF-1α) are critical in promoting tumor angiogenesis[ 104 ]. Under hypoxic conditions, HIF-1α and HIF-1β subunits form heterodimers that activate the transcription of many target genes to adapt to the hypoxic environment of human cancer cells[ 105 ].…”
