Selective serotonin reuptake inhibitor treatment retunes emotional valence in primate ventral striatum

Abstract: HAL is a multi-disciplinary open access archive for the deposit and dissemination of scientific research documents, whether they are published or not. The documents may come from teaching and research institutions in France or abroad, or from public or private research centers. L'archive ouverte pluridisciplinaire HAL, est destinée au dépôt et à la diffusion de documents scientifiques de niveau recherche, publiés ou non, émanant des établissements d'enseignement et de recherche français ou étrangers, des labor… Show more

“… 20 , 21 This effect, however, is context dependent. 22 In addition, activation of DRN 5-HT neurons promotes waiting for future reward. 23 , 24 , 25 This may reflect enhanced active persistence rather than passive behavioral inhibition.…”

“…This has been an obstacle in refining their use in psychiatry. In this issue of Neuropsychopharmacology, Pasquereau et al [3] demonstrate how fluoxetine treatment alters neuronal encoding of rewards and punishments by putative medium spiny neurons in the primate ventral striatum, a key node in the neural circuitry underlying motivated behavior [1,2,4]. Fluoxetine caused changes in neuronal encoding of reinforcer valence that coincided with changes in the monkeys' motivation to approach or avoid stimuli predictive of reward or punishment.…”

“…Moreover, while theories of serotonergic antidepressant mechanisms have been proposed at the cellular and psychological levels, it is difficult to unify them without a computational understanding of DRN and the 5-HT system. The therapeutic efficacy of both SSRIs and psychedelics has been linked to their effects on the activity of DRN neurons, 5-HT receptor availability, and neuroplasticity [219][220][221][222][223][224] , which may then manifest as adaptive psychological changes, such as reduced negativity biases and improved cognitive flexibility 219,[225][226][227][228][229] . The efficacy of these drugs in treating other psychiatric illnesses such as anxiety or obsessive-compulsive disorder may also depend on similar mechanisms [230][231][232][233][234][235][236][237] .…”
