Role of Matrix Metalloproteinases in Angiogenesis and Its Implications in Asthma

Abstract: Asthma is a chronic airway disorder associated with aberrant inflammatory and remodeling responses. Angiogenesis and associated vascular remodeling are one of the pathological hallmarks of asthma. The mechanisms underlying angiogenesis in asthmatic airways and its clinical relevance represent a relatively nascent field in asthma when compared to other airway remodeling features. Matrix metalloproteinases (MMPs) are proteases that play an important role in both physiological and pathological conditions. In addi… Show more

“…Myeloperoxidase (MPO), contained in neutrophil primary/azurophilic granules, plays a pivotal role in ROS production [72,73] and NET formation [74,75]. Matrix metalloproteinase-9 (MMP-9) plays a key role in the development of airway inflammation [76][77][78][79] and facilitates angiogenesis in asthma [80]. Circulating [81] and BAL levels of CXCL8 [82] are increased in asthmatics.…”

“…However, MMP-9 deficiency in mouse models led to increased interleukin-6 levels in the bone marrow, which promoted pancreatic ductal adenocarcinoma (PDAC) invasion and metastasis. Furthermore, VEGF may also serve as an driver because it can lead to upregulation of other pro-angiogenic factors ( 24 ). Vascular endothelial growth factor and MMP-9 are shown to be a key target in the “angiogenic on-off.”…”

“…EMT regulation by growth factors and others also showed that extrinsic factors, including IL-6, Tnf, TGF-b, Notch, Wnt, and receptor tyrosine kinase EGF, HGF, PDGF, FGF, etc., can further enhance cell invasion ability and facilitate cells to secrete matrix metalloproteinases (MMPs) to digest ECM [ 179 , 180 , 181 ]. Currently, approximately 23 MMP members have been identified [ 181 ].…”
