Nicotine Increases Spontaneous Glutamate Release in the Rostromedial Tegmental Nucleus

Abstract: The rostromedial tegmental nucleus (RMTg) is a bilateral structure localized in the brainstem and comprise of mainly GABAergic neurons. One of the main functions of the RMTg is to regulate the activity of dopamine neurons of the mesoaccumbens pathway. Therefore, the RMTg has been proposed as a modulator of the reward system and adaptive behaviors associated to reward learning. The RMTg receives an important glutamatergic input from the lateral habenula. Also, it receives cholinergic inputs from the laterodorsa… Show more

“…The summed activity was graphed below the raster plot in a form of a histogram (coactivity histogram) (Carrillo‐Reid et al., 2008). Monte Carlo simulations (1000) were used to find the significance of neurons being active together (Pérez‐Ortega et al., 2016; Lara‐González et al., 2019; Castillo‐Rolón et al., 2021). MATLAB was used to analyze the activity of individual cells along with the different conditions as described previously (Aparicio‐Juarez et al., 2017).…”

“…In contrast, amphetamine works by targeting the dopamine transporter, thereby increasing dopamine levels by transporter-mediated exchange (Rudnick and Clark, 1993). Glutamatergic systems have also been implicated in nicotine and amphetamine use liability behaviors (Castillo-Rolón et al, 2021;D'Souza and Markou, 2013;Palmatier et al, 2008). Glutamate plays an important role in learning and memory more generally (Zhou and Danbolt, 2014), but also as it relates to drug use behaviors (D'Souza and Markou, 2013).…”

“…Nicotine, the active ingredient in E‐cig, exerts its psychoactive effects by binding to nicotinic acetylcholine receptors (nACh receptors) in the brain. In the central nervous system (CNS), nACh receptors play crucial roles in mediating cholinergic transmission, neurotransmitter and growth factor release (Castillo‐Rolon et al, 2020; Serres & Carney, 2006). nACh receptors have been implicated in various CNS disorders, including TBI, Alzheimer's Disease (AD), Parkinson's Disease (PD) and other neurodegenerative conditions.…”
