A multidisciplinary investigation on Achillea moschata Wulfen (Asteraceae) is outlined herein. This work, part of the European Interreg Italy–Switzerland B-ICE project, originated from an ethnobotanical survey performed in Chiesa in Valmalenco (Sondrio, Lombardy, Northern Italy) in 2019–2021 which highlighted this species’ relevance of use in folk medicine to treat gastrointestinal diseases. In addition, this contribution included analyses of the: (a) phytochemical profile of the aqueous and methanolic extracts of the dried flower heads using LC-MS/MS; (b) morpho-anatomy and histochemistry of the vegetative and reproductive organs through Light, Fluorescence, and Scanning Electron Microscopy; (c) biological activity of the aqueous extract concerning the antioxidant and anti-inflammatory potential through cell-based in vitro models. A total of 31 compounds (5 phenolic acids, 13 flavonols, and 13 flavones) were detected, 28 of which included in both extracts. Covering and secreting trichomes were observed: the biseriate 10-celled glandular trichomes prevailing on the inflorescences represented the main sites of synthesis of the polyphenols and flavonoids detected in the extracts, along with volatile terpenoids. Finally, significant antioxidant and anti-inflammatory activities of the aqueous extract were documented, even at very low concentrations; for the first time, the in vitro tests allowed us to formulate hypotheses about the mechanism of action. This work brings an element of novelty due to the faithful reproduction of the traditional aqueous preparation and the combination of phytochemical and micromorphological research approaches.

Chemical Profiles of Korean Bee Pollens and Their Catechol-O-methyltransferase Inhibitory Activities

Bee pollen is an apicultural product collected by honeybees from flower stamens and is consumed to help maintain a healthy diet. In this study, the chemical profiles of 11 Korean bee pollens were investigated using molecular networking analysis. This analysis elucidated the presence of two major clusters, hydroxycinnamoyl acid amides (HCAAs, molecular network 1 (MN 1 )) and flavonoid glycosides (MN 2 ), in the bee pollen samples. The inhibitory properties of the bee pollens and the isolated HCAAs toward human catechol-O-methyltransferase (COMT), a key neurotransmitter involved in Parkinson's disease and depression, were determined. N 1 ,N 5 ,N 10 -(E)-tricaffeoylspermidine ((E,E,E)-1) exhibited the highest activity of the four compounds isolated, with an IC 50 value 16 μM, and inhibited COMT competitively. Quantitative analysis of HCAAs showed that the amounts of N 1 ,N 10dicaffeoyl-N 5 -p-coumaroylspermidine (2) and N 10 -caffeoyl-N 1 ,N 5 -di-p-coumaroylspermidine (3) contributed to the observed differences in the COMT inhibitory activities of Korean bee pollens. This study may lead to the prevention and treatment of Parkinson's disease and depression using bee pollens.

The purpose of this study was to prepare eight sub-fractions from leaves and flowers of Erica arborea L., characterize their phytochemicals constituents and investigate their potential antioxidant, in order to validate the beneficial medicinal properties of this shrub in Algeria folk medicine. Total polyphenols, flavonoids and condensed tannins contents were determined using Folin-Ciocalteu’s, aluminum chloride and vanillin reagents, respectively. The in vitro antioxidant activity was evaluated by using 2,2-diphenyl-l-picrylhydrazyl and reducing power assay. E. arborea L. contains various compounds such as polyphenols, flavonoids, tannins, quinones, anthraquinones, saponins and terpenoids in different sub-fractions. All the tested extracts showed an appreciable total phenolic, flavonoids and condensed tannins contents as well as strong antioxidant capacity. The highest total phenolic and flavonoids content was found in the ethyl acetate extracts ranging from 649.38 to 944.55 mg gallic acid equivalent/g dry extract and 65.31 to 67.15 mg quercetin equivalent/g dry extract, respectively. Whereas, the highest condensed tannins content was found in the crude extract for leaves and aqueous extracts for flowers. The ethyl acetate extract of the flowers and the crude extract of leaves exhibited the better antioxidant activity by DPPH assay (IC50 =17.72 µg/mL) and reducing power assays (IC50 = 2.91 µg/mL), respectively. Our findings indicate that leaves and flowers extracts are rich in natural antioxidant substances and have good qualities in antioxidant properties and may be beneficial against diver’s disorders related to free radicals.
