Structural Features of Amyloid Fibrils Formed from the Full-Length and Truncated Forms of Beta-2-Microglobulin Probed by Fluorescent Dye Thioflavin T

Abstract: The persistence of high concentrations of beta-2-microglobulin (β2M) in the blood of patients with acute renal failure leads to the development of the dialysis-related amyloidosis. This disease manifests in the deposition of amyloid fibrils formed from the various forms of β2M in the tissues and biological fluids of patients. In this paper, the amyloid fibrils formed from the full-length β2M (β2m) and its variants that lack the 6 and 10 N-terminal amino acids of the protein polypeptide chain (ΔN6β2m and ΔN10β2… Show more

“…For the preparation of amyloid fibrils from beta-2-microglobulin, the protocol described in the work [ 64 ] was used. The samples of recombinant beta-2-microglobulin were received from Mikhail M. Shavlovsky, Dmitry S. Polyakov, and Rodion G. Sakhabeev (Department of Molecular Genetics, Institute of Experimental Medicine).…”

“…Lysozyme (ALys amyloidosis) and beta-2-microglobulin (DRA), as proteins with significantly different secondary and tertiary structures and stability, were chosen by us as target proteins. Amyloid fibrils formed from these proteins were prepared by previously developed approaches [ 42 , 43 ].…”

“…To prepare amyloid fibrils from beta-2-microglobulin, we followed the special method used in the work [ 42 ]. The recombinant beta-2-microglobulin was kindly provided by Mikhail M. Shavlovsky, Dmitry S. Polyakov and Rodion G. Sakhabeev (Department of Molecular Genetics, Institute of Experimental Medicine).…”

“…Generally, we observed that Thioflavin T fluorescence reached a maximum, and then slowly declined. This pattern has been attributed to matting of fibrils, which can interfere with binding of the dye to fibrils, to selfquenching of ThT and related fluors, and to complexities of multiple aggregation products [42][43][44][45] . Fibrils obtained through successful seeding were examined by SSNMR (patients 1, 2, and 3) and XRD (the same three patients, and patient 4).…”
