Rapid diagnosis of Zika virus through saliva and urine by Loop-mediated isothermal amplification (LAMP)

Abstract: Background: Zika virus (ZIKV) is a single-stranded RNA virus and member of the Flaviviridae family. Recent studies have reported that saliva can be an important alternative to detect ZIKV. Saliva requires less processing than blood greatly simplifying the assay. Loop-mediated Isothermal Amplification (LAMP) is a rapid assay that detects nucleic acids, including ZIKV RNA. Aim: The aim of this study was to evaluate the efficacy of saliva and urine to diagnose ZIKV infection in subjects during the acute phase, th… Show more

“…Using 131 clinical samples from suspected patients collected during the acute phase of the disease, including 68 saliva and 63 urine specimens, an RT-LAMP assay was able to detect the ZIKV in 53.4% of the total number of samples. Interestingly, this study showed that ZIKV detection using saliva had a faster time to positivity compared to urine samples [61].…”

“…To provide perspective on the analytical sensitivity of different LAMP assays for ZIKV, we assembled a table summarizing this measure for all studies using ZIKV-spiked or RNA-spiked samples (Table 1) [27,28,[37][38][39][40][41][42]44,53,[57][58][59][60][61][62][63][64]. The top-performing ZIKV RT-LAMP method could detect ZIKV RNA standard at as few as 0.02 PFU/test, using RNA prepared by in vitro transcription.…”

“…[7,10]. Thus, LAMP could be a highly beneficial and convenient application to detect Zika virus due to its high specificity and stability, and it has been reported that LAMP shined a light on Zika virus [11][12][13][14][15][16][17][18][19].…”
