Histochemistry of the zona pellucida of the ovary of a species with natural polyovulation: Lagostomus maximus (Rodentia, Hystricomorpha, Chinchillidae)

Abstract: This study reports the histochemistry and the distribution of glycoconjugates (GCs) in the zona pellucida (ZP) of preantral, secondary, tertiary, polyovulatory and atretic follicles of ovaries from non-pregnant (NPr) and pregnant (Pr) females of Lagostomus maximus. GCs were studied using histochemical and lectin histochemical methods. The viscacha ZP was positive to all the histochemical techniques. In addition, it was observed that the intensity of staining of the ZP was constant in the different follicular s… Show more

“…These data indicate the presence of GlcNAc and β‐ d ‐galactose in these ovarian components of sows. Similar findings were noted in the zona pellucida of prepubertal and adult pigs (Pastor et al, ), viscacha (Acuña et al, ) and wild ungulates (Parillo, Dall’Aglio, Verini Supplizi, Ceccarelli, & Gargiulo, ). In addition, UEA‐1 presented weak or absent reaction in the zona pellucida in the present investigation, whereas other researchers detected mild (Parillo et al, ) or moderate (Pastor et al, ) marcation in the porcine zona pellucida.…”

“…Lectin histochemistry (LHC) is an important technique effectively used in the identification and in situ characterization of carbohydrates in ovarian follicles of many species, such as rat, mare, pig, viscacha ( Lagostomus maximus ) and women (Acuña et al, ; Barbeito et al, ; Desantis et al, ; Maymon et al, ; Pastor et al, ; Salvetti et al, ). This technique was also used to identify glycan residues in equine and porcine cumulus–oocyte complexes in in vitro ‐ and in vivo‐matured samples (Accogli et al, ).…”

“…Moreover, the WGA, DBA, SBA, RCA‐I and PNA lectins identified the glycosidic residues GlcNac and β‐galactose, and CON‐A identified mannose and glucose residues. In mammals, these residues have an important role in the oocyte‐sperm interaction (Acuña et al ., 2018; Amari et al ., 2001; Barbeito et al ., 2013; Maegawa et al ., 2002; Yonezawa et al ., 2005). However, in Chondrichthyes, fertilization occurs in the anterior oviduct (McMillan, 2007), so the same function is not possible.…”

“…For the identification of specific sugar residues (Goldestein & Hayes, 1978) in OG, a battery of seven biotinylated lectins was used (Table 1). For this, a standardized protocol from our laboratory was used (Acuña et al ., 2018). As negative controls, slices of oviductal gland incubated only with phosphate buffered saline were used.…”
