3'-Terminal sequences of the RNA genomes of narcissus latent and Maclura mosaic viruses suggest that they represent a new genus of the Potyviridae.

Abstract: The nucleotide sequences of part of the nuclear inclusion body b (NIb) gene, the complete coat protein gene and the 3h untranslated regions of narcissus latent virus (NLV) and Maclura mosaic virus (MacMV) were determined. Deduced amino acid sequences for the NIb and coat protein genes revealed that NLV and MacMV are closely related. Gel analysis and Western blotting of the coat proteins of NLV and MacMV from infected tissue or purified virus indicated that they have molecular masses of 39n5 kDa and 40 kDa (res… Show more

“…These data confirm the relationships of the macluraviruses and the ipomovirus within the family Potyviridae (Badge et al, 1997;Colinet et al, 1996). In this analysis the ipomovirus SPMMV is shown to be a sister taxon to SCSMV, WSMV, and BrSMV.…”

“…In this analysis, the Bymovirus genus is represented by BYMV and WSSMV. MacMV and NLV are members in a probable new genus Macluravirus (Badge et al, 1997). The Rymovirus representatives are WSMV, BrSMV, AgMV, HoMV, and RgMV and the Ipomovirus is SPMMV.…”

“…The western blot analysis of crude sorghum leaf extract with the antiserum of SStMV-AP clearly indicated the presence of a 40 kDa viral CP, an unusual size for most of the potyviruses. Coat proteins of the size of 39-40 kDa have been previously reported for viruses such as Narcissus latent virus, Maclura mosaic virus and SStMV-AP, although the deduced amino acid sequences of these viruses indicate a range of only 32-34 kDa (Badge et al 1997;Hema et al 1999). The M r of the virus studied herein, deduced from the CP (∼32 kDa) sequence, was also lower than the M r determined by western blot analysis (Fig.…”

“…These values are considerably less than the species demarcation threshold established by the International Committee on the Taxonomy of Viruses (ICTV). This degree of divergence among species within genera of the family Potyviridae is not uncommon [17][18][19][20]. In contrast, comparison of the WEqMV genome sequence with those of other potyvirids indicated that WEqMV is distantly related (B38.7% at nucleotide level and B20.7% at amino acid level) to rymoviruses, potyviruses, ipomoviruses, and bymoviruses.…”
