3-Methylcholanthrene Induces Chylous Ascites in TCDD-Inducible Poly-ADP-Ribose Polymerase (Tiparp) Knockout Mice

Abstract: TCDD-inducible poly-ADP-ribose polymerase (TIPARP) is an aryl hydrocarbon receptor (AHR) target gene that functions as part of a negative feedback loop to repress AHR activity. Tiparp−/− mice exhibit increased sensitivity to the toxicological effects of 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD), including lethal wasting syndrome. However, it is not known whether Tiparp−/− mice also exhibit increased sensitivity to other AHR ligands. In this study, we treated male Tiparp−/− or wild type (WT) mice with a single… Show more

“…The association between PARP10 and PCNA helps cells bypass stalls at replication forks and resume DNA synthesis. PARP7, however, has not been directly linked to DNA repair but does function in transcription regulation, cellular autophagy, immunity, and response to viral infections [12]. This review will focus primarily on PARP1 and PARP2 as most inhibitors act upon these enzymes.…”

“…CYP1A1, CYP1B1 and TIPARP are the target genes of AHR [ 46 , 47 ]. The mRNA levels of these target genes are significantly enhanced by I3P and I3A, which are inhibited by CH223191, a specific AHR inhibitor.…”

“…Tiparp −/− and Tiparp Hep −/− mice exhibit increased AHR activity and increased sensitivity to TCDD-induced toxicities and wasting syndrome ( Ahmed et al , 2015 ; Hutin et al , 2018 ). This increased sensitivity to AHR ligands is not limited to TCDD, since 3-methylcholanthrene treatment also causes severe toxicity and lethality to Tiparp −/− mice ( Cho et al , 2019 ). Overall, the TCDD-treated Tiparp H532A mice exhibited many, if not all, of the same phenotypes as similarly treated Tiparp −/− and Tiparp Hep −/− mice.…”
