MicroRNA-378 is involved in hedgehog-driven epithelial-to-mesenchymal transition in hepatocytes of regenerating liver

Abstract: Healthy livers have a remarkable regenerative capacity for reconstructing functional hepatic parenchyma after 70% partial hepatectomy (PH). Hepatocytes, usually quiescent in normal healthy livers, proliferate to compensate for hepatic loss after PH. However, the mechanism of hepatocyte involvement in liver regeneration remains unclear. Hedgehog (Hh) pathway plays an important role in tissue reconstitution by regulating epithelial-to-mesenchymal transition (EMT) in liver disease. MicroRNA (miRNA) is involved in… Show more

“…The most overexpressed miRNA in patients’ cells was the miR-378-3p, which is considered a modulator of the epithelial-to-mesenchymal transition and is associated with inflammation and fibrosis through the positive modulation of NF-kB and TNFα pathways [167,168]. miRNA-224, which is also up-regulated in hEDS/HSD cells, is associated with the activation of the Wnt/β-catenin signaling through the inhibition of the expression of glycogen synthase kinase 3β and SFRP2, which are known Wnt suppressors [169].…”

“…Activation of the HH pathway and increased c-Myc expression not only upregulated proliferation but also initiated EMT, which is important for cancer metastasis. The role of the HH pathway in EMT is well documented in cancer cells undergoing metastasis or tumor progression [75][76][77][78][79]. HH pathway signaling has been shown to have pathological effects in many cancer types [3].…”

“…It was known that circRNA played a role in post-transcriptional regulation by several ways such as miRNA sponges or protein templates [14]. After predicting circRNA-miRNA pairs, the predicted target miRNAs of circ_03848, including rno-miR-150-5p and rno-miR-378a-5p were reported to have a correlated expression pattern with it and associated with LR [34,35], suggesting that they could be directly bound to circ_03848 to affect rat LR. In detail, it was found that circ_03848 exhibited an upregulated pattern at 30 h after PH in this paper, and it might target rno-miR-150-5p and rno-miR-378a-5p.…”
